The Type VI Secretion System Plays a Role in Type 1 Fimbria Expression and Pathogenesis of an Avian Pathogenic Escherichia coli Strain

Abstract: Avian pathogenic Escherichia coli (APEC) strains frequently cause extraintestinal infections and are responsible for significant economic losses in the poultry industry worldwide. APEC isolates are closely related to human extraintestinal pathogenic E. coli (ExPEC) strains and may also act as pathogens for humans. Known APEC virulence factors include adhesins such as type 1 fimbriae and curli, iron acquisition systems, and cytotoxins. Here we show that APEC strain SEPT362, isolated from a septicemic hen, expre… Show more

“…Gene aec26 is chromosomally located and encodes a putative membrane protein component of a type VI secretion system (T6SS) (52,54). In an APEC strain, this T6SS has recently been shown to contribute to the adhesion and invasion of epithelial cells (HeLa) by influencing the expression of F1 fimbriae and to improve the virulence of the strain for 1-day-old chicks (15). O78 lipopolysaccharide contributes to serum resistance and colonization of internal organs of inoculated chicken with an APEC strain of serogroup O78 (40).…”

“…More recently, the translocation of canonical protein effectors degrading the peptidoglycan layer into the periplasm of recipient bacterial cells has been demonstrated (10,11). The T6SS has also been shown to be involved in stress sensing or biofilm formation, although the mechanistic bases for these phenotypes have not been described yet (12)(13)(14). The T6SS is therefore a versatile machine that adapts to the specific needs of each bacterium.…”

“…Flagellin contributes to persistence and invasion after oral dosing of chicks with APEC O78 (28), and ExPEC invasins (Ibe proteins) also facilitate APEC invasion (35,36). Indirect effects on the expression of virulence-associated fimbriae have been reported for mutants with a constitutively activated pho regulon (37) and those lacking the pst system (38), ibeA, ibeB, and ibeT (36,39), the barA-uvrY-encoded two-component sensory system (40), and a type VI protein secretion system (41). These reports may partially explain the attenuation of such mutants in chickens and indicate that the effect of mutations may propagate through complex networks.…”
