The tumor microenvironment and metastatic disease

Abstract: The microenvironment of solid tumors is a heterogeneous, complex milieu for tumor growth and survival that includes features such as acidic pH, low nutrient levels, elevated interstitial fluid pressure (IFP) and chronic and fluctuating levels of oxygenation that relate to the abnormal vascular network that exists in tumors. The metastatic potential of tumor cells is believed to be regulated by interactions between the tumor cells and their extracellular environment (extracellular matrix (ECM)). These interacti… Show more

“…Hypoxic environments are randomly scattered throughout carcinomas owing to the heterogeneous nature of angiogenesis [1]. Although hypoxia is cytotoxic to both cancer cells and normal cells, some types of cancer cells acquire characteristics that allow them to survive and progress in a hypoxic environment [2].…”

“…Zdolność ta umożliwia komórkom migrację i zasiedlanie zarówno okolicznych, jak i odległych tkanek [42]. Zjawisko przerzutowania stanowi kluczowy problem w leczeniu chorób nowotworowych, a jego wystąpienie przyczynia się do istotnego pogorszenia rokowania chorego [43].…”

“…This is clearly demonstrated when one alters the cellular environment to enable invasion and metastatic dissemination. For example, hypoxia (low oxygen) is known to be a potent driving force for metastatic progression, and there is a plethora of experimental studies to show that oxygen deprivation of cancer cells enhances their invasive and metastatic potential [35][36][37][38]. These studies have led to the identification of promising anti-metastatic targets, such as lysyl oxidase (LOX) [37,38].…”
