The Tumor-Associated Marker, PVRL4 (Nectin-4), Is the Epithelial Receptor for Morbilliviruses

Abstract: PVRL4 (nectin-4) was recently identified as the epithelial receptor for members of the Morbillivirus genus, including measles virus, canine distemper virus and peste des petits ruminants virus. Here, we describe the role of PVRL4 in morbillivirus pathogenesis and its promising use in cancer therapies. This discovery establishes a new paradigm for the spread of virus from lymphocytes to airway epithelial cells and its subsequent release into the environment. Measles virus vaccine strains have emerged as a promi… Show more

“…Alternatively, based on positive immunostaining, we speculate that CDV may also use N4 to breach the ependymal and meningeal barriers, which are, throughout the CNS, in intimate contact with an underlying astroglial cell layer (glia limitans). This view might be consistent with the reported frequent periventricular and subpial localization of CDV-induced demyelinating lesions (20,46,47) with intense infection of the ependymal cells. Once astrocytes are targeted, the infection will further spread deeply in the white matter in a GliaR-dependent manner.…”

“…Many of the PVR/PVRL family members have been shown to be involved in virus entry. PVR was first identified as the poliovirus receptor (39), followed by PVRL1 and PVRL2 as herpesvirus receptors (40,41), and most recently PVRL4 has been identified as an epithelial receptor for measles virus (42,43) and other morbilliviruses (44,45). This study represents the first report to our knowledge of a pathogen using PVRL3 as a receptor, and also the first instance to our knowledge of a bacterial toxin using a member of this family of proteins as a receptor.…”

“…More recently, nectin-4 was identified as an entry point for measles and other viruses and was proposed as a target for oncolytic viral therapy (21,22). Although several groups reported the detection of nectin-4 expression in breast, ovarian, and lung cancers (23)(24)(25)(26), no comprehensive analysis has been reported to date.…”
