The Transport of Prolamine RNAs to Prolamine Protein Bodies in Living Rice Endosperm Cells[W]

Abstract: RNAs that code for the major rice storage proteins are localized to specific subdomains of the cortical endoplasmic reticulum (ER) in developing endosperm. Prolamine RNAs are localized to the ER and delimit the prolamine intracisternal inclusion granules (PB-ER), whereas glutelin RNAs are targeted to the cisternal ER. To study the transport of prolamine RNAs to the surface of the prolamine protein bodies in living endosperm cells, we adapted a two-gene system consisting of green fluorescent protein (GFP) fused… Show more

“…Zein mRNAs could be actively translated and the resulting polypeptides immediately degraded, or the mRNAs could be translated at very low rates. Transcripts of prolamins in the rice (Oryza sativa) starchy endosperm have been shown to be recruited to specific ER domains where they are translated and the corresponding protein translocated to form to ER protein bodies (Choi et al, 2000;Hamada et al, 2003;Crofts et al, 2010). Although it is a matter of debate whether this is also the case for maize prolamin transcripts (Kim et al, 2002;Washida et al, 2004), proteins required for the efficient accumulation of zein aggregates inside the ER may not be expressed in aleurone cells.…”

“…We have argued that such PM-cER junctions are likely to exist in other eggs (for review, see Sardet et al, 2002). Several recent reports also indicate that specific cortical ER compartments close to the PM are present in yeast and plant cells, where they are thought to play important roles in polarization, membrane-protein insertion (in yeast bud) and mRNA and protein localization (in rice endosperm) (Du et al, 2004;Hamada et al, 2003).…”

“…Chloramphenicol inhibits elongation by plastid ribosomes. mRNAs may be sequestered into several types of non-translated particles, including transport granules (Hamada et al, 2003a), stress granules, and processing (P-) bodies. Stress granules form transiently during abiotic stress such as heat shock (Nover et al, 1989).…”
