The Transcriptional Signature of a Runner’s High

Abstract: Introduction Endorphins, endocannabinoids, monoamines, and neurotrophins have all been implicated in the euphoric response to endurance running, known as a runner’s high (RH). The epitranscriptional mechanisms regulating this effect have not been defined. Here, we investigate peripheral micro–ribonucleic acid (miRNA) changes unique to athletes experiencing postrun euphoria, yielding insights into gene networks that control an RH. Methods A cohort study … Show more

“…Starting with the influence on mood, various types of exercise, both acute and chronic, and ending with resistant and aerobic training, all activate the endocannabinoid signaling and result in significant mood improvements, antidepressant effect, reduced anger, and tension, as well as increased vigor and motivation [ 35 , 40 , 44 , 52 ]. Euphoric and analgesic phenomena widely described by athletes that occur during a forced and prolonged physical activity called “runner’s high” are the result of the activity of endorphins, monoamines, and endocannabinoids and their influence on the reward system in the brain [ 36 , 53 ]. Therefore, the “runner’s high” may lead to heavy exercise addiction probably, due to endogenous opioids release, which may be augmented by the ECS [ 54 , 55 ].…”

“…Endorphins and enkephalins play a role in motivational and stress circuits and have been implicated in neuroadaptations to drug abuse (Koob and Volkow, 2016). Both endorphin and enkephalin have profound pain-relieving effects and promote euphoria (Shenoy and Lui, 2018;Hicks et al, 2019).…”

“…Various human activities trigger the production of endorphins or enkephalins. Laughter is known to increased resistance to pain, stimulate endorphin production [ 158 ], and vigorous aerobic exercise releases β-endorphin [ 159 , 160 ]. Endorphins are released by pleasurable behaviors like eating chocolate, which contains TIQs, sex, orgasm, eating food, yoga, meditation, and listening to music [ 161 , 162 ].…”
