The Tomato Cf-2 Disease Resistance Locus Comprises Two Functional Genes Encoding Leucine-Rich Repeat Proteins

Abstract: In plants, resistance to pathogens is frequently determined by dominant resistance genes, whose products are proposed to recognize pathogen-encoded avirulence gene (Avr) products. The tomato resistance locus Cf-2 was isolated by positional cloning and found to contain two almost identical genes, each conferring resistance to isolates of tomato leaf mould (C. fulvum) expressing the corresponding Avr2 gene. The two Cf-2 genes encode protein products that differ from each other by only three amino acids and conta… Show more

Several R genes have been shown to belong to large, clustered gene families (Jones et al, 1994;Whitham et al, 1994;Lawrence et al, 1995;Dixon et al, 1996). However, the detailed genomic distribution of these multigene families is as yet not known.…”

“…Many of the cloned plant resistance genes show sequence homology, of which the most striking property is the occurrence of leucine-rich repeats in their gene products [11][12][13][14][15][16][17][18][19]. The only leucine-rich protein of which the crystal structure has been determined is porcine ribonuclease inhibitor [20][21][22].…”

“…M, the presumed homologous locus in this ancient tetraploid, has ∼86% nucleotide identity to L and comprises an array of ∼15 genes (paralogs); seven specificities are known at M . Occasionally two functional sequences encode the same specificity, as in the case of Cf2 (Dixon et al 1996). More usually only a single gene in the cluster determines resistance as shown by mutant or transgenic complementation analysis.…”
