The Tolerogenic Function of Regulatory T Cells in Pregnancy and Cancer

Abstract: Regulatory T cells, a subpopulation of suppressive T cells, are potent mediators of self-tolerance and essential for the suppression of triggered immune responses. The immune modulating capacity of these cells play a major role in both transplantation, autoimmune disease, allergy, cancer and pregnancy. During pregnancy, low numbers of regulatory T cells are associated with pregnancy failure and pregnancy complications such as pre-eclampsia. On the other hand, in cancer, low numbers of immunosuppressive T cells… Show more

“…However, there are some instances where CTLA-4 co-inhibitory signals are constitutively high and this can halt T cell activation (100). Such scenarios are seen in many cases of cancers and suppressor T cells such as T regulatory cells, suggesting the reason for lack of immune responses in cancer patients (101,102). CTLA-4 protein is homologous to CD28 protein and has a higher binding affinity toward CD80 and CD86 on major histocompatibility complex (MHC) (Figure 2).…”

“…More specifically, regulatory T cells (Treg cells) recognize male antigens and develop a tolerogenic immune environment by suppression of inflammation and immune rejection responses [100,101]. Treg cells are a population of T lymphocyte with immune suppressive properties that comprise both CD4+ and CD8+ subtypes [102]. Treg cells generally act to suppress cytokine synthesis and effector function in macrophages, T-, B-, natural killer (NK), and dendritic cells [103,104].…”

“…71 Increased regulatory T cells with a highly suppressive function CD4 + Tregs function primarily to suppress immune responses and thus maintain homeostasis and tolerance. 72 High tumor Treg infiltration has been reported in ovarian, liver, melanoma and oesophageal cancers, and is associated with an aggressive phenotype (extensive review by Jørgensen et al 73 ).…”
