The Telomerase-Derived Anticancer Peptide Vaccine GV1001 as an Extracellular Heat Shock Protein-Mediated Cell-Penetrating Peptide

Abstract: Cell-penetrating peptides (CPPs), which can facilitate the transport of molecular cargo across the plasma membrane, have become important tools in promoting the cellular delivery of macromolecules. GV1001, a peptide derived from a reverse-transcriptase subunit of telomerase (hTERT) and developed as a vaccine against various cancers, reportedly has unexpected CPP properties. Unlike typical CPPs, such as the HIV-1 TAT peptide, GV1001 enabled the cytosolic delivery of macromolecules such as proteins, DNA and siRN… Show more

“…Long-term therapy with this combination was very well tolerated and produced the highest anti-Aβ IgG titers compared with other regimens. 61 UB-311, a synthetic peptide used as Aβ vaccine, has been advanced into an ongoing phase 2 study in patients with mild 7 and moderate AD. In phase 1, it induced a 100% responder rate in patients with AD.…”

“…A special CPP, GV1001, a 16‐amino acid peptidase derived from human telomerase reverse transcriptase (hTERT), was reported in 2013 131 . Compared to other CPPs that pierce cell membranes by electrostatic interaction with proteoglycan, GV1001 can deliver macromolecules into the cell by forming complexes with extracellular heat shock proteins 90 and 70 131,132 . Its role as an anti‐cancer vaccine peptide has been reported in treating advanced pancreatic cancer, 133,134 non‐small cell lung cancer, 135 melanoma 136 and prostate cancer 137 .…”
