The targeted anti-oxidant MitoQ causes mitochondrial swelling and depolarization in kidney tissue

Abstract: Kidney proximal tubules (PTs) contain a high density of mitochondria, which are required to generate ATP to power solute transport. Mitochondrial dysfunction is implicated in the pathogenesis of numerous kidney diseases. Damaged mitochondria are thought to produce excess reactive oxygen species (ROS), which can lead to oxidative stress and activation of cell death pathways. MitoQ is a mitochondrial targeted anti‐oxidant that has shown promise in preclinical models of renal diseases. However, recent studies in … Show more

“…Besides normal tubular mitochondria, there are 2 special kinds of mitochondria generated by topological transformation: swollen and donut-shaped mitochondria. Various stresses can induce a shift from a tubular mitochondrial morphology to multiple round organelles with some or all swollen [22][23][24][25]. Mitochondrial swelling in relation to mitochondrial permeability transition pore (mPTP) opening has been observed, for example, during reoxygenation-reperfusion after hypoxia [26,27].…”

“…As a third option, mitoQ may have increased ROS production and oxidative stress due to its long alkyl linker group ( Fig. S1; Maroz et al 2009;Reily et al 2013;Trnka et al 2015;Yasui et al 2017;Gottwald et al 2018). This may have promoted the consumption of tocopherol and xanthophylls to be used as radical scavengers (Britton et al 2004;Panda and Cherian 2014).…”

“…decyl-TPP + or dTPP + ). However, this chain length seems to favor membrane permeability, inhibiting the electron transport chain (ETC) and inducing higher superoxide radical generation (see Reily et al 2013;Trnka et al 2015;Gottwald et al 2018). Interestingly, zebra finches only treated with dTPP + developed paler bills than controls (Cantarero and Alonso-Alvarez 2017).…”

“…This is likely to have improved the antioxidant action, avoiding disruption of the mitochondrial membrane and higher superoxide production (Reily et al . 2013; Trnka, Elkalaf & Anděl 2015; Gottwald et al . 2018).…”

“…The length of this molecule, however, increases membrane permeability, inhibiting the electron transport chain and rising superoxide radical generation (Reily et al . 2013; Trnka, Elkalaf & Anděl 2015; Gottwald et al . 2018).…”
