Cadmium (Cd) pollution poses potential safety risks for Panax notoginseng cultivation, a medicinal plant in Yunnan. Under exogenous Cd stress, field experiments were conducted to understand the effects of lime (0, 750, 2250 and 3750 kg hm−2) applied and oxalic acid (0, 0.1 and 0.2 mol L−1) leaves sprayed on Cd accumulation, antioxidant system and medicinal components of P. notoginseng. The results showed that Lime and foliar spray of oxalic acid were able to elevate Ca2+ and alleviate Cd2+ toxicity in P. notoginseng under Cd stress. The addition of lime and oxalic acid increased the activities of antioxidant enzymes and alters osmoregulator metabolism. The most significant increase in CAT activities increased by 2.77 folds. And the highest increase of SOD activities was 1.78 folds under the application of oxalic acid. While MDA content decreased by 58.38%. There were very significant correlation with soluble sugar, free amino acid, proline and soluble protein. Lime and oxalic acid were able to increase calcium ions (Ca2+), decrease Cd content and improve the stress resistance of P. notoginseng, while increasing the production of total saponins and flavonoids. Cd content were the lowest, 68.57% lower than controls, and met the standard value (Cd ≤ 0.5 mg kg−1, GB/T 19086-2008). The proportion of SPN was 7.73%, which reached the highest level of all treatments, the flavonoids content increased significantly by 21.74%, which reached the medicinal standard value and optimal yield.

Background The optimization of process parameters in the pharmaceutical industry is often carried out according to the Quality by Design (QbD) concept. QbD also emphasizes that continuous improvement should be performed in life cycle management. Process parameters that are difficult to control in actual production could be regarded as noise parameters. In this study, a noise parameter was considered, an example of continuous improvement in the design space was provided. Methods The ethanol precipitation process of Carthami Flos (Honghua) extract was optimized based on the QbD concept. The critical process parameters (CPPs) were identified using a definitive screening design. Considering that the refrigeration temperature of industrial ethanol precipitation is often difficult to control with seasonal changes, the refrigeration temperature was treated as a noise parameter. The design space was then calculated using an exhaustive search-Monte Carlo method. The mathematical models were reestablished when more data were obtained and then the calculated probabilities of reaching the process standards were updated. Results The calculation procedure of design space based on an exhaustive search-Monte Carlo method was proposed. The density of the concentrated extract, ethanol concentration, the volume ratio of ethanol to concentrated extract, stirring time after ethanol addition, and refrigeration temperature were selected as CPPs. The mathematical models of CPPs and evaluation indicators were established, and the coefficient of determination of each model was greater than 0.81. The predictive performance of the models was good. After continuous improvement, the recalculated probability values were more reliable, the design space became larger. Conclusions The calculation of design space and the continuous improvement strategy considering a noise parameter was developed. In industrial production, it is also recommended to adopt this similar idea, that is, continuing to collect industrial data and regularly updating the mathematical models, which can further update the design space and make it more stable and reliable.

BACKGROUND A comprehensive literature search shows that Sanqi and Huangjing (SQHJ) can improve diabetes treatment in vivo and in vitro , respectively. However, the combined effects of SQHJ on diabetes mellitus (DM) are still unclear. AIM To explore the potential mechanism of Panax notoginseng (Sanqi in Chinese) and Polygonati Rhizoma (Huangjing in Chinese) for the treatment of DM using network pharmacology. METHODS The active components of SQHJ and targets were predicted and screened by network pharmacology through oral bioavailability and drug-likeness filtration using the Traditional Chinese Medicine Systems Pharmacology Analysis Platform database. The potential targets for the treatment of DM were identified according to the DisGeNET database. A comparative analysis was performed to investigate the overlapping genes between active component targets and DM treatment-related targets. We constructed networks of the active component-target and target pathways of SQHJ using Cytoscape software and then analyzed the gene functions. Using the STRING database to perform an interaction analysis among overlapping genes and a topological analysis, the interactions between potential targets were identified. Gene Ontology (GO) function analyses and Kyoto Encyclopedia of Genes and Genomes enrichment analyses were conducted in DAVID. RESULTS We screened 18 active components from 157 SQHJ components, 187 potential targets for active components and 115 overlapping genes for active components and DM. The network pharmacology analysis revealed that quercetin, beta-sitosterol, baicalein, etc. were the major active components. The mechanism underlying the SQHJ intervention effects in DM may involve nine core targets (TP53, AKT1, CASP3, TNF, interleukin-6, PTGS2, MMP9, JUN, and MAPK1). The screening and enrichment analysis revealed that the treatment of DM using SQHJ primarily involved 16 GO enriched terms and 13 related pathways. CONCLUSION SQHJ treatment for DM targets TP53, AKT1, CASP3, and TNF and participates in pathways in leishmaniasis and cancer.
