The structure of helix 89 of 23S rRNA is important for peptidyl transferase function of Escherichia coli ribosome

Abstract: Edited by Michael IbbaKeywords: Ribosome Peptidyltransferase rRNA Mutagenesis a b s t r a c t Helix 89 of the 23S rRNA connects ribosomal peptidyltransferase center and elongation factor binding site. Secondary structure of helix 89 determined by X-ray structural analysis involves less base pairs then could be drawn for the helix of the same primary structure. It can be that alternative secondary structure might be realized at some stage of translation. Here by means of site-directed mutagenesis we stabilized … Show more

“…Finally, the CTD occupies the A site of the 50S subunit, and its β-turn (residues 527-532) interacts with H89 (Fig. S4E), which is an important element of ribosome because it connects the PTC and the elongation factor binding site (33). Furthermore, the distal loop residues (552-555 and beyond 555) appear to touch the hairpin loop of H92 at the PTC.…”

“…The helix H89 resides at the 60S subunit surface facing the 40S subunit and extends from the PTC ring (Anger et al 2013). According to numerous data obtained with bacterial ribosomes (e.g., see O'Connor and Dahlberg 1995;La Teana et al 2001;Burakovsky et al 2011), H89 is involved in different steps of the translation process. In eukaryotes, this helix is implicated in interaction of the ribosomes with eIF5B (Unbehaun et al 2007).…”
