The structure and properties of histone F2a comprising the heterologous group F2a1and F2a2studied by13C nuclear magnetic resonance

Abstract: (13)C n.m.r. spectra have been obtained for aqueous solutions of histones F2a(1) and F2a(2), for the group F2a, for the appropriate amino acid mixturesand for the corresponding hydrolysates. These, when compared with computer simulated spectra give good agreement for secondary structure with that calculated from the known primary structure of the proteins. Evidence based on the spectra obtained at various salt concentrations leads to the conclusion that F2a is not a simple mixture but an interacting heterologo… Show more

“…This is in keeping with our previous finding (1) that both pairs are required together with DNA to generate the x-ray diffraction pattern characteristic of chromatin. It is also in keeping with the work of D'Anna and Isenberg (20) and Clark et al (21) but not with the suggestion of Hyde and Walker (22) that the pairs F2A1,FS and F2A2,F2B do not interact.…”

“…Mobile C a carbons are consistent only with random coil peptides; ' (iii) Relative mobility should be deduced from relaxation times. Previous n.m.r, investigations [4,16,23,24] of histones have relied upon differences in linewidth to estimate mobilities. This cannot be used as a rigorous test of backbone mobility since observed resonances from histones are generally composed of a number of partially overlapping resonances and hence v½ does not reflect 7"2 alone.…”

“…Histones interact with each other and with DNA [1][2][3][4][5][6] ;both kinds of association are essential for the integrity of chromatin. Current models for the structure of chromatin are based upon a periodic repeating unit in which a segment of DNA comprising about 140-160 base pairs is folded with a packing ratio of 7 or less into a 'bead' containing eight histone molecules, two each of F2A1, F3, F2A2 and F2B [7,8].…”

“…It has been possible both to determine which regions of the histones are involved in homologous interactions and also to show that the aggregation process does not affect the secondary structure of the histones. The original I3C studies of histones F2Al and F2A2 (Clark et al, 1974) have been extended and similar studies made with histones F2B and F3.…”
