The skin barrier state of aged hairless mice in a dry environment

Abstract: We confirmed that a dry environment induces epidermal proliferation and scaling in both aged and young mice. However, no remarkable difference was found in the skin barrier recovery of aged hairless mice in a dry environment.

“…The only significant difference detected was the TEWL between ESS with mDPC-GFP and host murine skin. Very low TEWL in skin of athymic mice has been reported previously [15]. These results suggested no compromise of skin integrity in either of the ESS models.…”

“…Epidermal Ca 2+ has been shown to coordinately regulate both lamellar body secretion and terminal differentiation (i.e. cornified envelope formation, loss of cellular organelles and synthesis of differentiation‐specific proteins) 29–31 . In addition, epidermal terminal differentiation is also directed by activation of caspase 14, a cysteinyl aspartate‐specific proteinase (reviewed in Denecker et al.…”

[29][30][31] In addition, epidermal terminal differentiation is also directed by activation of caspase 14, a cysteinyl aspartate-specific proteinase (reviewed in Denecker et al 32 ). To determine which processes are controlled specifically by ER Ca 2+ emptying, we treated normal hairless mice with a single topical application of a low concentration (100 nmol L )1 ) of TG, applied to unperturbed flank skin.…”

“…Stratum corneum hydration and TEWL were measured using Corneometer CM825 and Tewameter TM300 (Courage and Khazaka Electronics, Cologne, Germany) at 3 weeks and 6 weeks. 27,28) Probes were applied to the skin surfaces of mice. After the 6-week feeding period, mice were deprived of food overnight and anesthetized with diethyl ether.…”
