The simultaneous role of porphyrins’ H- and J- aggregates and host–guest chemistry on the fabrication of reversible Dextran-PMMA polymersome

Abstract: Aggregation-induced quenching of porphyrin molecules as photosensitizer significantly reduces the quantum yield of the singlet oxygen generation, and it is able to decrease the efficacy of photodynamic therapy. We utilized amphiphilic copolymers in this work to precisely control porphyrin H-type and J-type aggregations in water. The amphiphilic copolymer bearing azobenzene, β-cyclodextrin, and porphyrin was successfully synthesized by the atom transfer radical polymerization technique. The azobenzene and β-cyc… Show more

“…UV-Vis spectroscopy and chromatography can, thus, be employed as a quantification strategy according to the cargo characteristics to quantify the cargo encapsulation efficiency (EE) and loading capacity (LC) [ 58 ]. Alternatively, UV-vis spectroscopy has straightforwardly monitored the photocleavage [ 80 ] and photoisomerization [ 200 ] properties in light-triggered polymersomes by evaluating the spectra evolutions upon light irradiation.…”

“…The dual-drug encapsulation efficiency was 32.6 for Q and 37.3% for 5-FU. The results also showed that the 5-FU release was higher than Q due to its hydrophilic character [ 200 ].…”

“…By encapsulating (a part of) the photosensitizer in the cavities, its self-aggregation was suppressed to accomplish a high quantum yield of 1 O 2 production. The aggregation of porphyrin was photocontrolled by using complex formation of β-CyD with photoresponsive azobenzene [ 78 ].…”

“…This then results in poor solubility in aqueous environments, a rapid clearance from the blood circulation, and therefore a low bioavailability. Additionally, the high aromaticity results in undesired aggregation, leading to a quenching of the singlet oxygen generation and thereby to a reduced therapeutic effect [ 18 ]. An important parameter to evaluate the effect of PSs is the phototoxic index (p.i.…”
