The serological pattern of autoantibodies to the Ro52, Ro60, and La48 autoantigens in primary Sjögren's syndrome patients and healthy controls

Abstract: We found a distinct and significant difference in the serum antibody specificity and immunoglobulin isotype pattern between SS patients and matched controls. This variance may point to different mechanisms by which these autoantibodies are generated.

“…Another prominent feature of SS is the high production of autoantibodies against the intracellular proteins Ro/SSA and La/SSB, which can be detected in both the peripheral blood [20][21][22] and SG [23][24][25] of SS patients. Recently, it was reported that most primary SS patients produce autoantibodies many years before symptom onset [26,27].…”

“…The autoantibody concentrations were generally higher in sera from Sjo¨gren's syndrome patients and posses a more heterogeneous autoantibody repertoire then healthy subjects with regard to the number of autoantigens and antibody isotypes (Garberg et al, 2005).…”

“…Ro, a ribonucleoprotein to which autoantibodies can be detected in up to 75% of human patients with SS [4], has a role in the etiology and pathology of SS as yet uncertain. Studies have shown that reciprocal spreading to the Ro52, Ro60, and La polypeptides occurs after immunization with a single component.…”

“…Histopathologically, SS is characterized by progressive focal mononuclear cell infiltration of the exocrine glands [2], destruction and/or functional defect [3], leading to clinical symptoms of dry mouth and dry eyes (xerostomia and keratoconjunctivitits sicca, respectively). Patients frequently present with autoantibodies to intracellular ribonucleoproteins such as Ro/SSA and/or La/SSB [4] and rheumatoid factor (RF) [1].…”
