“…Most of the HPLC methods previously used to assay clozapine and its metabolites in serum/plasma, have used chemically modified silica L ---(bonded phase) column packing materials and aqueous methanol or acetonitrile eluents most commonly in the pH range 2-5. As an alternative, we chose to investigate the use of unmodified (Jane, 1975;Lingeman and Undderberg, 1988;Binder et al, 1989;Law, 1990;Flanagan, 1993) and sulphopropyl modified silica (McCarthy et al, 1993;Croes et al, 1994a,b) systems with methanolic eluents containing ammonium perchlorate (pH6.7) or perchloric acid (0.01-1.0% vlv) as modifiers with the aim of injecting sample extracts directly onto the column. As a means of further varying the flexibility of the system, two chemically bonded stationary phases, cyanopropyl-modified and phenyl- modified silica were also tried with these eluents (Flanagan, 1993).…”

“…Methanol solutions of the compounds were autoinjected by a Perkin-EImer ISS-100 automatic sampler. A modified Wheals system [14,15] was used as the mobile phase for each of the columns listed in Table 4. The ultraviolet (UV) detector was set at 254/~, a commonly used wavelength in fixed wavelength detectors.…”

“…Hydrophilic interaction chromatography (HILIC) using bare silica or derivatized silica and low aqueous/high organic mobile phase has drawn the attention of LC-MS/MS practitioners. Although the name HILIC was coined by Alpert in 1990 on a derivatized silica column [2], this approach was first used by Jane in 1975 on an underivatized silica column [3]. HILIC is similar to normal phase LC in that the elution is promoted by the use of polar mobile phase.…”
