The CLAVATA signaling pathway mediating stem cell fate in shoot meristems requires Ca2+ as a secondary cytosolic messenger

Abstract: , and that this cytosolic secondary messenger is involved in the signal transduction cascade linking CLV3/CLV1 to control of gene expression and stem cell fate in the SAM.

“…Because the isolated, enriched shoot apex tissues were used in this study but not whole seedlings, the use of different tissues may lead to the different results in the clv1 mutant. In addition, CLV1 has been reported to be expressed outside the SAM (Chou et al 2016). Based on our observation of MAPK activation, these results showed that CLV3p-mediated phosphorylation of MPK3 and MPK6 is positively regulated by both CLV1 and BAM1 receptors.…”

“…WUS and FAF2 expression is inhibited by the CLV3 peptide in wild-type plants and impaired with Ca 2+ channel blockers and GC inhibitors. This inhibition was also observed in the DEFENSE NO DEATH 1 ( dnd1 ) and 2 ( dnd2 ) mutants that lack the CNGC 2 and 4, respectively ( Chou et al, 2016 ). This may suggest that when CLV3 binds to the CLV1 receptor in the meristem, an increase in cGMP results and that may in turn activate CNGC2 channels and consequently increase cytoplasmic Ca 2+ .…”

“…Peptides are typically released as pro-peptides into the apoplast and, once cleaved, bind to LRR kinases. Interestingly, 3 pathways that are initiated by peptides (CLAVATA3 (CLV3), phytosulfokine (PSK) and peptide PEP3) use cGMP as a secondary messenger to link with downstream signals (Qi et al 2010;Kwezi et al 2011;Chou et al 2016). Several of these receptors contain a GC domain within their kinase domain (i.e PEPR1, CLV1).…”
