The scavenger endothelial cell: a new player in homeostasis and immunity

Abstract: To maintain homeostasis, the animal body is equipped with a powerful system to remove circulating waste. This review presents evidence that the scavenger endothelial cell (SEC) is responsible for the clearance of blood-borne waste macromolecules in vertebrates. SECs express pattern-recognition endocytosis receptors (mannose and scavenger receptors), and in mammals, the endocytic Fc gamma-receptor IIb2. This cell type has an endocytic machinery capable of super-efficient uptake and degradation of physiological … Show more

“…It is generally accepted that extracellular macromolecules, as well as entire cells, are turned over by uptake and lysosomal degradation in specialized scavenger cells, namely, macrophages and scavenger endothelial cells (SECs). 35 Functionally, macrophages represent the professional phagocyte responsible for the elimination of larger particles. 35 While clearance of the BsAbs via resident liver macrophages was plausible, the lack of effect of macrophage depletion on clearance (Fig.…”

“…Noteworthy, HSPGs are required for the initial recruitment of Tat onto the cell surface, and a scavenger receptor low-density lipoprotein receptor-related protein is responsible for the exact uptake of Tat (53). Importantly, LL-37 is recognized as a CPP (36), and LSECs express various scavenger receptors (54). Thus, it is reasonable to speculate that the LL-37-LPS complex initially interacts with cell-surface HSPGs and subsequently binds with some scavenger receptor(s) on LSECs for the incorporation.…”

“…It has been shown that endothelial cells-especially hepatic sinusoidal endothelial cells, a representative RES member-comprise an important part of the innate immune system for eliminating foreign particles from the circulation. Smedsrød and his colleagues (32,36,37) have termed the endocytic endothelial cells as scavenger endothelial cells, leading us to resurrect the concept of the RES (39). A weak point in this effort to re-evaluate the RES is the lack of marker substances that can specify the RES members.…”
