The roles of the inhibitory autophagy regulator Rubicon in the heart: A new therapeutic target to prevent cardiac cell death

Abstract: Autophagy contributes to the maintenance of cardiac homeostasis. The level of autophagy is dynamically altered in heart disease. Although autophagy is a promising therapeutic target, only a few selective autophagy activator candidates have been reported thus far. Rubicon is one of the few endogenous negative regulators of autophagy and a potential target for autophagy-inducing therapeutics. Rubicon was initially identified as a component of the Class III PI3K complex, and it has multiple functions, not only in… Show more

“…According to the NCCD, autophagic cell death is a regulated form of cell death that relies on autophagy machinery and can be prevented only by blocking autophagy 2 . Autophagic cell death has been implicated in various physiological and pathological processes, including cancer, neurodegeneration, ischemic injury, and heart disease 70 74 . However, the precise role of autophagy in cell death is still unclear 16 .…”

“…8 Furthermore, Rubicon negatively regulates autophagosome-lysosome fusion by interacting with Beclin-1. 121 It is reported that CXCL10 blocked the fusion of autophagic vesicles with lysosomes through regulating Rubicon. Moreover, genetic inhibition of Rubicon reversed bile acid that induced incomplete autophagy.…”

“…They demonstrated that the SARS‐CoV‐2‐ORF3a‐UVRAG interaction renders the binding of UVRAG to Beclin‐1 lower competitive than that of ATG14, changes the balance between Beclin‐1‐VPS34‐UVRAG and Beclin‐1‐VPS34‐ATG14 complex, thus promoting more efficient formation of autophagosomes but inhibiting maturation of autophagosome 8 . “… 7 Autophagy contributes to the maintenance of cardiac homeostasis, and the level of autophagy is dynamically altered in heart disease. 8 , 9 Mitophagy, which mediates the selective elimination of dysfunctional mitochondria, is essential for cardiac homeostasis. 10 In cardiomyocytes, mitophagy is closely associated with metabolic activity, cell differentiation, apoptosis, and other physiological processes involved in major phenotypic alterations.…”
