The roles of plant dsRNA‐binding proteins in RNAi‐like pathways

Abstract: Dicers are associated with double-stranded RNAbinding proteins (dsRBPs) in animals. In the plant, Arabidopsis, there are four dicer-like (DCL) proteins and five potential dsRBPs. These DCLs act redundantly and hierarchically. However, we show there is little or no redundancy or hierarchy amongst the DRBs in their DCL interactions. DCL1 operates exclusively with DRB1 to produce micro (mi)RNAs, DCL4 operates exclusively with DRB4 to produce trans-acting (ta) siRNAs and 21nt siRNAs from viral RNA. DCL2 and DCL3 p… Show more

“…The biased accumulation of miRNA duplex strands was also assessed in our dsRBP combinational insertion mutant knockout line, drb2345 (Curtin et al 2008). In this line, only one, DRB1, of the five known Arabidopsis dsRBPs is functional.…”

“…Furthermore, DCL4 exclusively interacts with DRB4, to efficiently process both viral and trans-acting siRNAs from their dsRNA substrates (Hiraguri et al 2005;Adenot et al 2006;Nakazawa et al 2007;Curtin et al 2008). Therefore, we studied the accumulation of these two miRNAs and their respective miRNA* sequences in drb1 and drb4 mutant plant lines (Fig.…”

“…DCLs associate with dsRNA-binding proteins (DRB) which assist in the production of siRNAs [73]. In Arabidopsis, DRB4 operates with DCL4 to produce siRNAs from viral RNA [30]. The interaction of CDL4 and DRB4 in viral siRNA production has been shown in experiments both in vitro and in vivo [3].…”

“…DCLs' cofactors, the DRB proteins are also required for vsiRNA biogenesis. DRB4, the cofactor of DCL4, takes part in antiviral defense against RNA viruses (Curtin et al 2008;Jakubiec et al 2012;Qu et al 2008 silencing suppressor of Cauliflower mosaic virus (CaMV) inhibits DRB4, strongly suggests that DRB4 is an antiviral factor against DNA viruses as well. DRB3, the cofactor of DCL3, contributes to antiviral defense through chromatin modification against DNA viruses (Raja et al 2014).…”
