The Roles of Non-coding RNA in the Development and Regeneration of Hair Follicles: Current Status and Further Perspectives

Abstract: Alopecia is a common problem that affects almost every age group and is considered to be an issue for cosmetic or psychiatric reasons. The loss of hair follicles (HFs) and hair caused by alopecia impairs self-esteem, thermoregulation, tactile sensation and protection from ultraviolet light. One strategy to solve this problem is HF regeneration. Many signalling pathways and molecules participate in the morphology and regeneration of HF, such as Wnt/β-catenin, Sonic hedgehog, bone morphogenetic protein and Notch… Show more

“…Epithelial cell differentiation and hair pigmentation occur during this stage. The duration of the anagen stage determines hair length and is associated with the continuous proliferation and differentiation of stromal cells at the base of hair follicles [ 11 , 13 , 14 ]. Moreover, there is substantial variation in anagen duration according to body region: up to 6 years in the scalp, 1 to 3 months in the arms, 4 to 6 months in the legs, and 1 to 2 months in the thighs.…”

“…When hair follicles enter the catagen stage, hair usually stops growing and, at the end of this phase, the hair follicles have atrophied. Therefore, the catagen stage starts when the supply of matrix cells declines, leading to a slowdown in the differentiation of the hair shaft and the inner root sheath (apoptosis-driven regression) [ 13 , 14 ]. During catagen, hair follicle degeneration is highly regulated, and a large number of keratinocytes from the hair follicle begin to undergo programmed death.…”

“…Unlike messenger RNAs (mRNAs), which do not encode proteins, ncRNAs are ubiquitous throughout the human genome ( Yang et al, 2021b ). ncRNAs play an essential regulatory role in various biological processes such as cell proliferation, epigenetic modification, and cell apoptosis ( Yang et al, 2021b ). The top three most commonly described ncRNA molecules are miRNAs, long ncRNAs (lncRNAs), and circular RNAs (circRNAs) ( Smith et al, 2021 ).…”

“…The short non-coding sub-type microRNAs (miRNAs) are implicated in the temporal- and spatial-specific regulation of all body organs and tissues, including skin morphogenesis and hair follicle cycling and growth [ 8 , 9 ]. Recent evidence highlighted the importance of exploring this type of non-coding RNA to unravel the underlying pathophysiological mechanisms of AA and to discover novel biomarkers that identify its severity, differentiate its subtypes, or predict the therapeutic response [ 10 , 11 , 12 , 13 ].…”
