The Role of Translationally Controlled Tumor Protein in Tumor Growth and Metastasis of Colon Adenocarcinoma Cells

Abstract: Translationally controlled tumor protein (TCTP) plays a major role in a broad array of biological processes. However, the TCTP-related biological process and interactive proteins still remain poorly characterized. In the present study, we found that knockdown of TCTP inhibited proliferation, migration, and invasion activities of LoVo cells in vitro and in vivo. The whole-cell proteomes were compared by 2D gel electrophoresis before and after knockdown of TCTP. Alterations in 27 proteins were detected and their… Show more

“…Another proteomics study on colon cancer cells identified 27 proteins, whose levels were altered after TCTP knockdown. In particular, components of ubiquitin-proteasome system and proteins involved in the cytoskeleton were affected under this condition (Ma et al 2010). …”

“…TCTP protein levels are upregulated in cancer cells and in human tumours (Tuynder et al, 2004;Kim et al, 2008a;Ma et al, 2009;Slaby et al, 2009). Downregulation of TCTP has been implicated in biological models of tumour reversion (Tuynder et al, 2002(Tuynder et al, , 2004, and the protein is the target of various anticancer drugs (Tuynder et al, 2004;Efferth, 2005), of which one has entered a phase-I/II clinical trial (Telerman and Amson, 2009).…”

“…22,23 In addition, TCTP has been suggested as a predictive marker for colon adenocarcinoma progression and tumor metastasis. 24 The increased expression of TCTP in FTC versus FA was confirmed by Western blot analysis (Figure 4). Furthermore, the protein DJ-1 was significantly up-regulated in FTC compared with ST (2.5-fold; P Ͻ 0.02; Table 1).…”
