The role of toll-like receptors (TLRs) in urinary tract infections (UTIs)

Abstract: IntroductionUrinary Tract Infections (UTIs) are caused by different types of microbial agents such as uropathogenic Escherichia coli (UPEC) and Candida albicans. The presence of strong physical barriers may prevent the breach of pathogens into the urinary tract. However, sometimes the pathogenic microorganisms may pass through the barriers and stimulate the innate and adaptive responses. Among a variety of innate immune responses, Toll-Like Receptors (TLRs) are one of the most unique and interesting molecules … Show more

“…The type 1 pilus is 2 µm in length and 10 nm in width, and is highly represented in the bacterial surface (100-500 pili per cell) [5,[47][48][49]. This pilus is defined as mannose-sensitive, because it is able to interact with the mannosylated receptors expressed by epithelial cells, particularly urothelial cells [12,19,48,53,54]. This specific function relies on the expression of the adhesin FimH located at the tip of the type 1 pilus.…”

“…They are expressed highly in bladder cells but have low expression in kidney cells. 47 As shown in previous studies, Tlr5-/-mice are more susceptible to UPEC infection than wild-type ones. 48 Preventing flagellin-induced signalling, the TLR5_C1174T polymorphism was associated with a higher risk of recurrent UTI in a cohort of 987 women.…”

“…Inherent immune recognition and regulation are mainly realized by a series of embryo-coded pattern recognition receptor (PRR) to recognize those pathogens associated molecular pattern (PAMP) expressed on the pathogenic microorganisms. Toll-like receptors (TLRs) are important pattern recognition receptor families in natural and acquired immunity, and act as pivotal priming proteins for mammals to transmit extracellular antigen signal information into cells [9]. TLR can initiate a series of signal transduction pathways, activate the effect factors in the pathway, amplify the cascade responses of downstream signal pathway, cause the aggregation of granulocytes, macrophages and natural killer cells, increase vascular permeability, stimulate the release of downstream in ammatory mediators, cause slight in ammatory reactions in the body, and nally activate the immune regulation function of the body to achieve the effect of controlling and eliminating pathogens.…”
