The Dapper/Frodo family of proteins are Dishevelled-interacting regulators of Wnt signaling. In this study, I characterize the regulation of the early expression patterns of dpr1 and dpr2. Although both dpr1 and dpr2 are expressed on the prospective dorsal side, I find that their pregastrula expression patterns have differences that have not been reported previously. Early dpr1 expression is much more dynamic than dpr2 expression. I use gain and loss of function experiments to identify dorsal organizer genes that regulate dpr1 and dpr2 expression. The dorsalizing factors ␤-catenin, Bozozok (Boz), Noggin (Nog), and the mesendoderminducing factor Squint (Sqt) are all able to induce ectopic expression of dpr1 and dpr2. In reciprocal loss of function experiments, loss of maternal ␤-catenin signaling leads to loss of early dorsal dpr1 and dpr2 expression, whereas loss of Boz and/or Nodal signaling does not. Ectopic expression of the ventralizing molecule Bmp2b leads to reduction of dpr1 and dpr2 expression. These results suggest that, in early zebrafish development, dpr1 and dpr2 are targets of ␤-catenin and/or an unknown downstream effector.

During vertebrate embryogenesis, endoderm, the inner‐most germ layer, will form the gut and associated organs, including liver and pancreas. Studies in the zebrafish, Xenopus and mouse have shown that the signalling pathways and transcription factors that regulate endodermal cell fate in the early embryo are largely conserved in vertebrates. Evidence from these model systems indicates Nodal signalling induces endoderm, in combination with the activity of transcription factors from the Mix, Gata, Sox and T‐box families. Other signalling pathways, such as Fgf, Bmp and Wnt, ensure the correct balance of mesoderm and endoderm is formed during gastrulation. Key Concepts Endoderm formation is initiated during mid‐blastula stages in zebrafish and Xenopus . High levels of Nodal signalling induce endoderm formation. A conserved set of transcription factors, of the Sox, Mix, Gata and T‐box families, act in a network with Nodal to establish endodermal fate during gastrulation. The role of these factors and pathways downstream of Nodal signalling in early endoderm formation is mostly conserved; however, there are some species‐specific differences. Other signalling pathways, notably the Fgf, Bmp and Wnt pathways, regulate the amount of endoderm that forms versus other germ layers.

Taken together, each teratogen produced a unique set of phenotypic changes in the body axis, suggesting that each compound affects Shh-s and also produces a distinctive set of molecular alterations. However, addition of exogenous Shh to ethanol treated zebrafish prevented many of the gross physical phenotypes, suggesting that the suppression of Shh-s is one of the major effects of ethanol exposure.
