The Role of the REPLUMLESS Homeodomain Protein in Patterning the Arabidopsis Fruit

Abstract: The outside of the Arabidopsis thaliana fruit consists of three principal tissues: the valves or seedpod walls, the replum or central ridge between the valves, and the valve margins where the valves separate from the replum to disperse the seeds. Previous studies have shown that valve margin formation is specified by the SHATTERPROOF MADS-box transcription factors and that valve development is controlled by the FRUITFULL MADS-box transcription factor. FRUITFULL negatively regulates SHATTERPROOF to prevent the … Show more

“…In carpels, KNAT6 and KNAT2 are expressed in the boundaries between the replum and the valves, while BP, STM, and PNY are expressed in the replum (this work; Long et al, 1996;Roeder et al, 2003;Alonso-Cantabrana et al, 2007). Both its layered structure and its role in generating ovule primordia make the replum a true meristem.…”

“…pny mutants are characterized by phyllotaxy defects, partial loss of apical dominance, and reduced stature (Byrne et al, 2003;Roeder et al, 2003;Smith and Hake, 2003;Bhatt et al, 2004). In addition, the pny mutant shows fruit alterations, such as defects in replum differentiation and in septum fusion (Byrne et al, 2003;Roeder et al, 2003). To investigate the interaction between KNAT2, KNAT6, and PNY, we generated the knat2 pny, knat6 pny, and knat2 knat6 pny mutants.…”

“…Valve margin identity is developed and maintained via the expression of several transcription factors including the MADS-box genes SHATTERPROOF1 (SHP1), and SHP2, and basic helix-loop-helix genes INDEHISCENT (IND) and ALCATRAZ (ALC) [80][81][82]. The activity of these regulatory genes is restricted to the valve margins, as their expression is repressed by REPLUMLESS (RPL) gene in the replum [83], and FRUITFULL (FUL) gene in the valves [84].…”
