IntroductionInterleukin‐38 (IL‐38) is a new type of anti‐inflammatory cytokine, which is mainly expressed in the immunity‐related organs and is involved in various diseases including cardiovascular and cerebrovascular diseases, lung diseases, viral infectious diseases and autoimmune diseases.AimThis review aims to detail the biological function, receptors and signaling of IL‐38, which highlights its therapeutic potential in related diseases.ConclusionThis article provides a comprehensive review of the association between interleukin‐38 and related diseases, using interleukin‐38 as a keyword and searching the relevant literature through Pubmed and Web of science up to July 2023.

Comprehensive analysis of characteristic genes of inflammation-related bronchopulmonary dysplasia based on bioinformatics methods

Background: Bronchopulmonary dysplasia (BPD) is the most common complication of premature infants, and inflammation plays an important role in it. In this research, the key genes of inflammation-related BPD were comprehensively analyzed by bioinformatics methods, and a prediction model was constructed. Methods: We obtained the GSE188944 and GSE108794 datasets from the GEO database and conducted GO and KEGG enrichment analyses to identify differential genes associated with inflammation-related BPD. Through the implementation of LASSO regression, RF, and XGBOOST algorithms, identify the key genes. The accuracy of these key genes in predicting BPD was assessed using ROC curve analysis and AUC calculations. In addition, the GSE190215 data set is used for external verification. Furthermore, we performed GSEA to quantify the key genes and analyze pathways, examine immune cell infiltration in BPD tissues, and explore the correlation amongst the key genes. Additionally, we used relevant databases to predict the miRNA and transcription factors associated with the key genes. Results: This study successfully identified seven key genes (HLA-DRB1, SLC39A8, IL2RA, SYK, CD180, IL6ST, IL18R1) as novel markers for constructing a diagnostic prediction model for BPD. GSEA analysis revealed enrichment pathways related to the key genes, and significant differences in the infiltration of CD8+ T cells, natural killer cells, and mast cells were observed between BPD and non-BPD samples. Conclusion: We successfully developed a risk model for inflammation-related BPD key genes, displaying favorable verification performance, but poor external verification performance. These findings suggest that inflammatory genes may influence the occurrence and progression of BPD through immune cell infiltration.

Influence of Umbilical Cord Blood Biochemical Parameters and Disease Condition on the Expression of the TSG-6 Gene in Umbilical Mesenchymal Stem Cells

Background Mesenchymal stem cells (MSCs) are capable of secreting different substances, including the anti-inflammatory protein TSG-6, which can be useful in the treatment of diseases with inflammatory reactions. The main aim of this study was to evaluate the expression of the TSG-6 gene in MSCs derived from the umbilical cord. For better understanding of the anti-inflammatory properties of MSCs, we additionally assessed the expression of some interleukins (ILs). Material/Methods The study group included 45 patients after delivery, aged from 21 to 46 years; the average patient age was 33 years. MSCs were isolated enzymatically from umbilical cord Wharton’s jelly, in vitro cultured, and characterized using flow cytometry; qPCR was performed to assess expression of the studied genes. The expression of genes of a number of pro-inflammatory ILs in MSCs was investigated in relation to the health of patients (coexistence of hypertension), the level of leukocytes, pCO2, and hemoglobin in the blood. Results Our research showed that the expression of the TSG-6 gene in MSCs depends on coexisting diseases in the patient and the biochemical parameters of umbilical cord blood, including the important role of cord blood pH. We found that the levels of IL2 and IL6 expression were correlated with pCO2, and IL6 expression were correlated with pO2. Conclusions Our study suggests that maternal health status and cord blood biochemical parameters could affect the anti-inflammatory properties of MSCs; however, this needs to be confirmed in a future study.
