The Role of Snail in EMT and Tumorigenesis

Abstract: Epithelial-mesenchymal transition (EMT) is a highly conserved process in which polarized, immotile epithelial cells lose adherent and tight junctions, and become migratory mesenchymal cells. As a key transcriptional repressor of E-cadherin expression in EMT, Snail plays an important role in embryonic development and cancer progression. Emerging evidences indicate that Snail confers tumor cells with cancer stem cell-like traits, and promotes drug resistance, tumor recurrence and metastasis. In this review, we s… Show more

“…TWIST overexpression promotes invasion of glioma cells in vitro and in orthotopic glioma xenotransplants in vivo by inducing the expression of EMT-associated genes like MMP-2 and FN-1. The SNAIL family of transcriptional repressors consisting of SNAIL/SNAI1 and SLUG/SNAI2 is known to drive invasion and metastasis in various carcinomas (163). SNAIL binds to E-box DNA sequences of genes related to an epithelial phenotype through carboxy-terminal zinc-finger domains, thereby suppressing their expression.…”

“…This process is followed by the downregulation of E-cadherin (37), which is considered to mediate homotypic cell-cell adhesion and maintain normal morphology, epithelial cell polarity, and tissue structural integrity by binding to cytosolic β-catenin and forming the E-cadherin/β-catenin complex. β-Catenin is required for the EMT as it binds to E-cadherin and functions as a key molecule in the Wnt signaling pathway (37). The Wnt signaling pathway is crucial for stem cell survival, proliferation, differentiation, and chemo-/radioresistance (37,38) due to the action of Wnt inhibitory factor 1 (WIF-1), a critical inhibitor of the Wnt signaling pathway (39).…”

The Wnt signaling pathway is crucial for stem cell survival, proliferation, differentiation, and chemo-/radioresistance (37,38) due to the action of Wnt inhibitory factor 1 (WIF-1), a critical inhibitor of the Wnt signaling pathway (39). Several studies have demonstrated that aberrant activation of the Wnt signaling pathway due to genetic and epigenetic alterations occurs in several types of cancer (39)(40)(41)(42).…”
