The Role of RelA and SpoT on ppGpp Production, Stress Response, Growth Regulation, and Pathogenicity in Xanthomonas campestris pv. campestris

Abstract: Xanthomonas campestris pv. campestris (Xcc) is a typical seedborne phytopathogenic bacterium that causes large economic losses worldwide, and this is the first original research article to investigate the role of ppGpp in this important species. Here, we revealed the function of RelA and SpoT in ppGpp production, physiology, pathogenicity, and stress resistance in Xcc.

“…Most of these 11 genes are involved in central metabolic pathways and were also found to be important for growth in artificial media in vitro (Table S2). Nevertheless, we detected (p)ppGpp metabolism as specifically important in xylem sap, suggesting that Xcc faces nutrient scarce conditions that require a stringent response (Irving et al, 2021; Bai et al, 2022).…”

“…However, it is interesting to note that the relA gene ( XC_1173 ) involved in (p)ppGpp biosynthesis was only found to be important for growth in xylem sap. Since accumulation of the (p)ppGpp alarmone triggers the stringent response and allows bacterial cells to face nutrient starvation, it is likely that Xcc feeds on anything available in the xylem to ensure multiplication (Bai et al, 2022). In the future, investigating the metabolite composition of guttation fluids and xylem sap will be needed to understand what is available for Xcc in the plant.…”

“…This is in agreement with the reduced biofilm formation on glass reported for V. cholerae [ 61 ] and Xanthomonas campestris pv. Campestris 8004 [ 122 ] mutants. Similarly, enhanced biofilms on PS were noticed in P. putida KT2440 [ 123 ] and V. alginolyticus [ 26 ].…”

“…and, therefore, facilitating bacterial pathogeny and dissemination [ 124 , 125 ]. Moreover, some authors have related it with the expression of various EPS operons (i.e., vps , pea , peb , bcs ) in diverse bacterial genera, such as Vibrio , Xanthomonas and Pseudomonas ; and, in consequence, with biofilm matrix production [ 26 , 61 , 122 , 123 ]. Moreover, the lack of spoT has also been related to low bacterial growth and no flagella formation, and, consequently, a reduced ability to form biofilms on glass tubes [ 61 , 126 ], which is consistent with our results on reinforced glass.…”

“…Most of these 11 genes are involved in central metabolic pathways and are important for growth in synthetic media (Table S3). (p)ppGpp metabolism was specifically important in xylem sap, where Xcc probably faces nutrient-scarce conditions requiring a stringent response (Irving et al, 2021;Bai et al, 2022). Before reaching the xylem, Xcc infects hydathodes where it resides as a biotroph for 3 d and becomes a necrotroph at 6 dpi (Cerutti et al, 2017).…”
