The Role of Nuclear Factor Kappa B in Lens Epithelial Cell Proliferation Using a Capsular Bag Model

Abstract: Purpose: This study was performed to elucidate the role of nuclear factor ĸB (NF-ĸB) in lens epithelial cell proliferation in the capsular bag model for this study. Methods: Capsular bags were prepared from porcine eyes and cultured in a 5% CO2 atmosphere at 37°C. NF-ĸB translocation was confirmed by immunohistochemistry and Western blot analysis. The effects of sulfasalazine and SN50 peptide, inhibitors of NF-ĸB activation, were observed by light microscopy and scanning electron microscopy. Results… Show more

“…This process was named pyroptosis [41,42]. It has been reported that NF-κB inhibits the proliferation and modulates the progression of lens epithelial cells [49]. In hippocampal neurons, treatment with TLR4 antibody, bpV(pic), or PDTC decreased the LPS-induced nuclear translocation of NF-κB and resulted in the down-regulation of TNF-α and IL-1β expression [50], which revealed that the TLR4-mediated NF-κB signaling pathway is associated with the activation of a neuroinflammatory response.…”

“…Further, in a recent study redox-sensitive transcription factor NF-κB has been implicated in LEC proliferation and PCO 25. We have shown that aldose reductase (AR), a polyol pathway enzyme that reduces glucose and various lipid peroxidation-derived aldehydes and their glutathione conjugates, mediates ROS signals initiated by growth factors, cytokines, high glucose and bacterial endotoxin (Lypopolysaccheride; LPS) via activation of NF-κB 26-29.…”

“…76 The sealed-capsule irrigation system is a device that has been developed to selectively deliver an agent to target lens epithelial cells following cataract removal. 77 The agents/mechanisms currently investigated to prevent PCO include nuclear factor kappaB (NF-kappaB), 78 proteasome inhibition, 79 macrophage depletion 80 as well as drugs like cyclosporin A 81 and mitomycin-C. 82 Moving away from the surgical options, drugs have been tried as a therapeutic option for cataracts. N-acetyl carnosine is one of the topical agents that have been developed for this purpose.…”
