The Role of NQO1 in Ovarian Cancer

Abstract: Ovarian cancer is one of the most dangerous gynecologic malignancies showing a high fatality rate because of late diagnosis and relapse occurrence due to chemoresistance onset. Several researchers reported that oxidative stress plays a key role in ovarian cancer occurrence, growth and development. The NAD(P)H:quinone oxidoreductase 1 (NQO1) is an antioxidant enzyme that, using NADH or NADPH as substrates to reduce quinones to hydroquinones, avoids the formation of the highly reactive semiquinones, then protect… Show more

“…CAP, a natural small molecule derived from chili peppers, has been identified and verified to modulate the KEAP1-NRF2-ARE pathway in our study. This signaling plays a key role in several cancerous and non-cancerous diseases [33][34][35][36][37] . Therefore, it is a promising therapeutic target for combatting free radical damage associated with numerous conditions, including Alzheimer's disease 38 , chronic kidney disease 39 , cardiovascular disease 40 , cancer and aging 41 .…”

“…Moreover, with each successive chemotherapy treatment and increased dosage, the intervals between treatments shorten, leading to heightened chemoresistance and a deteriorating prognosis 5 . Research suggests that heightened antioxidant capacities within cancer cells and increased capabilities to expel drugs are primary contributors to chemoresistance 6–9 . Additionally, an enhanced ability to repair DNA damage and the inactivation of tumor cell apoptosis are other prevalent factors 10,11 .…”

“…From a molecular standpoint, the increased antioxidant capacity of the cancer cells and the surrounding microenvironment was the primary cause of chemoresistance in ovarian cancer cells 9,10 . NQO1 is an antioxidant enzyme that takes part in ovarian cancer onset and progression 10 . Its modulator, Nrf2 mediates antioxidant pathway and serves as a resistance mechanism in proteasome inhibitor‐resistant cells 11 .…”
