We investigated the expression of components of the renin-angiotensin system (RAS) by cancer stem cell (CSC) subpopulations in metastatic head and neck cutaneous squamous cell carcinoma (mHNcSCC). Immunohistochemical staining demonstrated expression of prorenin receptor (PRR), angiotensin-converting enzyme (ACE), and angiotensin II receptor 2 (AT2R) in all cases and angiotensinogen in 14 cases; however, renin and ACE2 were not detected in any of the 20 mHNcSCC tissue samples. Western blotting showed protein expression of angiotensinogen in all six mHNcSCC tissue samples, but in none of the four mHNcSCC-derived primary cell lines, while PRR was detected in the four cell lines only. RT-qPCR confirmed transcripts of angiotensinogen, PRR, ACE, and angiotensin II receptor 1 (AT1R), but not renin or AT2R in all four mHNcSCC tissue samples and all four mHNcSCC-derived primary cell lines, while ACE2 was expressed in the tissue samples only. Double immunohistochemical staining on two of the mHNcSCC tissue samples showed expression of angiotensinogen by the SOX2+ CSCs within the tumor nests (TNs), and immunofluorescence showed expression of PRR and AT2R by the SOX2+ CSCs within the TNs and the peritumoral stroma (PTS). ACE was expressed on the endothelium of the tumor microvessels within the PTS. We demonstrated expression of angiotensinogen by CSCs within the TNs, PRR, and AT2R by the CSCs within the TNs and the PTS, in addition to ACE on the endothelium of tumor microvessels in mHNcSCC.

Mast cells (MCs) have important immunoregulatory roles in skin inflammation. Annexin A1 (ANXA1) is an endogenous anti-inflammatory protein that can be expressed by mast cells, neutrophils, eosinophils, monocytes, epithelial and T cells. This study investigated MCs heterogeneity and ANXA1 expression in human dermatoses with special emphasis in leprosy. Sixty one skin biopsies from 2 groups were investigated: 40 newly diagnosed untreated leprosy patients (18 reaction-free, 11 type 1 reaction/T1R, 11 type 2 reaction/T2R); 21 patients with other dermatoses. Tryptase/try+ and chymase/chy + phenotypic markers and toluidine blue stained intact/degranulated MC counts/mm were evaluated. Try/chy MCs and ANXA1 were identified by streptavidin-biotin-peroxidase immunostaining and density was reported. In leprosy, degranulated MCs outnumbered intact ones regardless of the leprosy form (from tuberculoid/TT to lepromatous/LL), leprosy reactions (reactional/reaction-free) and type of reaction (T1R/T2R). Compared to other dermatoses, leprosy skin lesions showed lower numbers of degranulated and intact MCs. Try MCs outnumbered chy in leprosy lesions (reaction-free/reactional, particularly in T2R), but not in other dermatoses. Compared to other dermatoses, ANXA1 expression, which is also expressed in mast cells, was higher in the epidermis of leprosy skin lesions, independently of reactional episode. In leprosy, higher MC degranulation and differential expression of try/chy subsets independent of leprosy type and reaction suggest that the Mycobacterium leprae infection itself dictates the inflammatory MCs activation in skin lesions. Higher expression of ANXA1 in leprosy suggests its potential anti-inflammatory role to maintain homeostasis preventing tissue and nerve damage.

Objective: To evaluate the expression of E-cadherin, Vimentin, CD31, CD117 in Oral Epithelial Dysplasia (OED) and Oral Squamous Cell Carcinoma (OSCC), in comparison with Normal Oral Mucosa (NOM). Material and Methods:Descriptive case study. Histological samples were analyzed by immunohistochemistry against E-cadherin, Vimentin, CD31, CD117. E-cadherin expression was evaluated by extent and intensity, Vimentin was evaluated by intensity and CD31 and C117 quantitatively. Results:The extent of E-cadherin was greater than 75% in all NOM samples, 81% of the OED samples and 47% of the OSCC samples. The intensity of E-cadherin was high in NOM (100%) and in OSCC (36%). Mean CD31 was 21.2 ± 5 in NOM samples, 23.4 ± 5.8 in OED samples and 36.7 ± 10.8 in OSCC samples. Mean CD117 was 7.2 ± 2.5 in NOM samples, 12.8 ± 5.3 in OED samples and 22.7 ± 5.3 in OSCC samples. Conclusion:The molecular marker E-cadherin is significantly more downregulated in dysplastic cells in comparison to NOM cells, while the mesenchymal marker Vimentin is expressed by epithelial cells in samples of NOM, OED and OSCC; questioning their value as prognostic markers. Finally, the quantification of CD31 and CD117 increased significantly in OSCC samples.
