The Role of KEAP1-NRF2 System in Atopic Dermatitis and Psoriasis

Abstract: The Kelch-like erythroid cell-derived protein with cap‘n’collar homology-associated protein 1 (KEAP1)-nuclear factor erythroid-2-related factor 2 (NRF2) system, a thiol-based sensor-effector apparatus, exerts antioxidative and anti-inflammatory effects and maintains skin homeostasis. Thus, NRF2 activation appears to be a promising treatment option for various skin diseases. However, NRF2-mediated defense responses may deteriorate skin inflammation in a context-dependent manner. Atopic dermatitis (AD) and psori… Show more

“…Nrf2 prevents or protects against oxidative stress-mediated cellular damage and promotes the expressions of antioxidant-related factors, such as HO-1 [ 46 ]. Moreover, the Nrf2/HO-1 signaling pathway provides a defense mechanism that regulates chronic inflammatory responses induced by oxidative stress [ 47 , 48 , 49 ]. Previous studies suggest that the activation of the Nrf2/HO-1 pathway enhances the inflammatory response and epidermal homeostasis from oxidative stress in hapten-induced AD mice [ 47 , 50 ].…”

Previous studies suggest that the activation of the Nrf2/HO-1 pathway enhances the inflammatory response and epidermal homeostasis from oxidative stress in hapten-induced AD mice [ 47 , 50 ]. In our study, the topical application of NPR extract increased the expressions of the antioxidant enzymes pNrf2, Nrf2, and HO-1, which regulate DNCB-induced excessive oxidative stress.…”

“…62 Activation of Nrf2 may improve inflammatory signalling in human keratinocytes in vitro and the phenotype of sensitizer-induced skin inflammation in AD mouse models. 63 NLRP3 inflammasome plays a vital role in the development of AD, and the activation of NLRP3 inflammasome induces AD. 42 Many studies have proven that suppressing NLRP3 inflammasome contributes to the mitigation of AD.…”

“…This results in a significant resistance at cellular levels to a subsequent highly lethal dose of oxidants. Recently, Ogawa and Ishitsuka [ 275 ] reviewed the role of Keap1-Nrf2 system in the pathophysiology of atopic dermatitis and psoriasis and the therapeutic approaches that utilize this system.…”
