The role of interleukin-24 in atopic dermatitis

Abstract: Atopic dermatitis (AD) is characterized by skin barrier disruption, type 2 immune dysregulation, chronic pruritus, and abnormal colonization by Staphylococcus aureus (S. aureus). Tapinarof, an aryl hydrocarbon receptor modulator, has been demonstrated to attenuate the development of AD in clinical studies. Recently, we found that tapinarof upregulated the expression of filaggrin and loricrin, which are essential proteins in skin barrier functions. Paradoxically, tapinarof induced interleukin (IL)-24 secretion … Show more

“…In POAD, the serum levels of IL‐24, significantly correlated with the expression of many inflammatory skin biomarkers, including Th17/Th22‐ (PI3/Elafin, KYNU, CXCL2, IL‐36A, DEFB4, KRT16, CCL7, S100A8/12), Th2‐ (OX40, CCR7, CCR4, TSLPR), innate immune‐ (IL‐8, IL‐6, IL‐1B), and Th1‐ (CXCL9/10/11, STAT1, CCL5) related markers (Figure 6D and Figure S8C,D). IL‐24 has been reported as upregulated in AD skin and was suggested to amplify skin inflammation, barrier disruption, and itch 50,51 …”

“…Skin immune diseases such as AD and allergies are caused by excessive skin inflammation. Since keratinocytes activated by various stimuli play an important role in the inflammatory immune response [ 19 ], we examined the anti-inflammatory effects of RCE on HaCaT cells under TNF-α and IFN-γ-induced inflammatory conditions.…”

“…Interleukin (IL) inhibitors are effective against CISD, owing to the involvement of IL family of cytokines in these conditions when dysregulated [399]. Ustekinumab, Secukinumab, Ixekizumab, Guselkumab, and Risankizumab are some examples, which act on targets known as IL proteins such as IL-1,12, 17, 23, and 24, hence decreasing the affinity to their natural receptors [400,401].
