The role of interleukin 22 in multiple sclerosis and its association with c-Maf and AHR

Abstract: The aim of this paper was to summarise knowledge of IL-22 involvement in multiple sclerosis (MS) and the possible link between IL-22 and two transcription factors-AHR and c-Maf. The conclusion is that despite numerous studies, the exact role of IL-22 in the pathogenesis of MS is still unknown. The expression and function of c-Maf in MS have not been studied. It seems that the functions of c-Maf and AHR are at least partly connected with IL-22, as both directly or indirectly influence the regulation of IL-22 ex… Show more

“…The role of IL-22, a proinflammatory member of the IL-10 cytokine family, in the pathogenesis of multiple sclerosis remains to date ambiguous [ 115 ]. It can act as a pathological agent in several autoimmunological diseases and exert a tumor-promoting effect in numerous cancer types, i.e., breast, colon and lung cancers [ 116 ]. It is highly expressed by Th17 cells; however, IL-22 can additionally be secreted by Th22, a fairly newly discovered T lymphocyte CD4+ subset which resides in tissues [ 117 ].…”

“…Apart from IL-17, Th17-like cells may also produce IL-22 (Zarobkiewicz et al 2019c), a cytokine acting mostly on non-hematological cells, e.g., epithelial cells (Rutz et al 2013). Under the majority of experimental settings, IL-22 was found to be protective in murine model of asthma; though in fungal asthma, it was found to be rather pathogenic (Hirose et al 2018).…”

“…Erk (extracellular-signal regulated kinase) participates in immunomodulation and has been proposed as a possible therapeutic target [119]. AHR (aryl hydrocarbon receptor) has recently been associated with interleukin-22, whose exact role in the pathogenesis of MS is still unknown [120]. Sod (superoxide dismutase) is a detoxicant enzyme that becomes important in the case of dysfunctional and impaired ROS metabolism occurring in neurodegenerative disorders such as MS [121].…”
