The Role of Host CD4 T Cells in the Pathogenesis of the Chronic Graft-versus-Host Model of Systemic Lupus Erythematosus

Abstract: Systemic lupus erythematosus is characterized by production of autoantibodies and glomerulonephritis. The murine chronic graft-vs-host (cGVH) model of systemic lupus erythematosus is induced by allorecognition of foreign MHC class II determinants. Previous studies have shown that cGVH could not be induced in CD4 knockout (CD4KO) mice. We have further explored the role of host CD4 T cells in this model. Our studies now show that B cells in CD4KO mice have intrinsic defects that prevent them from responding to a… Show more

“…The latter mechanism would be parallel to what we have found in CD4 Ϫ/Ϫ mice, in which the development of B cells in the absence of the CD4 T cell subset results in their inability to respond in the cGVH (19,21). In addition, several groups have reported a failure to detect Mer on lymphocytes (7,10).…”

“…Donor B cells disappear shortly after transfer and are not stimulated by the host T cells. Recipient B cells undergo polyclonal activation, accompanied by B cell phenotypic changes and increases in total serum levels of Igs (IgM and IgG), followed by the appearance of specific autoantibodies (19). We had predicted that Mer Ϫ/Ϫ mice would develop more rapid or robust cGVH, given their susceptibility to the spontaneous loss of tolerance presumably because of defective clearance of apoptotic cells (9).…”

“…cGVH was induced as previously described (21). In brief, disease was induced in 6-wk-old recipient mice by an intraperitoneal injection of a single cell suspension in HBSS of 10 8 donor spleen cells, prepared by pressing spleens through a wire mesh screen.…”
