The role of heat shock proteins 27 and 70 in redox-dependent regulation of apoptosis in Jurkat tumor cells

Abstract: Heat shock proteins Hsp) act as molecular chaperones, protecting enzymes and other proteins against reactive oxygen species. The objective of the study was to investigate the role of Hsp27 in maintaining the balance of the glutathione system and Hsp70 concentrations as well as in implementing Jurkat tumor cell apoptosis. Addition of the Hsp27 inhibitor KRIBB3 (5-(5-ethyl-2-hydroxy-4-methoxyphenyl)-4-(4-methoxyphenyl)-isoxazol) to Jurkat cells resulted in glutathione redox imbalance (increased GSSG and increase… Show more

“…It is worth mentioning that HSPH1 , HSPA6 , HSP70.1 , LOC102178315 (heat shock 70 kDa protein 1B), and HSPB1 were also highly expressed during catagen. These genes belong to the heat shock protein family and are involved in stress resistance, such as ensuring the correct folding of proteins or controlling the targeting of proteins for subsequent degradation [ 49 51 ]. These proteins would therefore be useful in preventing apoptosis, making a response to DPs adjacent organization.…”

“…It has been speculated that Hsp27 enhances the ability of migration and invasion of cancer cells by preventing dendritic cell maturation [ 6 ]. Blocking the expression of Hsp27 protein has become an important research direction in tumor targeted therapy [ 7 , 8 ]. The over-expression of Hsp27 has been detected in invasive tumor cells in breast carcinoma, oral squamous cell carcinoma, and pancreatic carcinoma [ 9 11 ].…”

“…Hsp27 is an ATP-dependent chaperone that folds substrate proteins and participates in shuttling destabilized or misfolded proteins toward proteolytic degradations [ 35 ]. Hsp27 is cytoprotective; it blocks apoptosis through interaction with various factors active in the apoptotic pathway [ 36 ]. The inhibition of apoptosis is required in the heat shock response (HSR) for cells to survive transient stress-induced apoptosis accompanied by the Hsp-mediated refolding of damaged proteins [ 37 ] ( Figure 2 ).…”
