The role of H2‐O and HLA‐DO in major histocompatibility complex class Il‐restricted antigen processing and presentation

Abstract: The function of major histocompatibility complex (MHC) class II molecules is to sample exogenous antigens for presentation to CD4+ T helper cells. After synthesis in the endoplasmic reticulum, class II molecules are directed into the endosomal system by association with the invariant chain (Ii), which is sequentially cleaved, generating class II dimers loaded with Ii-derived peptides (CLIP). These class II-peptide complexes are physiological substrates for H2-M/HLA-DM, a resident of the endosomal/lysosomal sys… Show more

“…Furthermore, the DO␤ promoter may serve as a model for identifying new transcriptional activators, transcriptional repressors, and/or chromatin remodeling factors involved in MHC class II gene activation. The DO heterodimer is also expressed in some, but not all, cultured dendritic cells (15,20,21), and it would be of interest to determine the parameters that determine dendritic cell expression. Lastly, it would be of interest to determine whether CIITA is involved in the LAN-5 neuroblastoma model in which DO␤ is induced by IFN-␥ (55).…”

“…In order for loading of peptides derived from antigens to occur, MHCII-bound CLIP must be exchanged for self- and foreign-derived peptides. Two MHCII homologues, H2-M (HLA-DM in humans; DM) and H2-O (HLA-DO in humans; DO) control this process (2, 3). H2-M facilitates the exchange of CLIP for antigen-derived peptides, prolongs the half-life of peptide receptive MHCII molecules and also ensures that only high affinity MHCII-peptide complexes are presented at the cell surface (48).…”

“…Peptide loading of class II molecules requires productive interactions with H-2M and H-2O molecules (44,45). Therefore, we determined whether the HLA class II molecules expressed in mouse cells formed stable dimers in SDS, indicating their assembly with peptide Ag.…”
