The role of FoxO1 and its modulation with small molecules in the development of diabetes mellitus: A review

Abstract: Diabetes mellitus type 2 (T2D) is one of the metabolic disorders suffered by a global human being. Certain factors, such as lifestyle and heredity, can increase a person's tendency for T2D. Various genes and proteins play a role in the development of insulin resistance and ultimately diabetes in which one central protein that is discussed in this review is FoxO1. In this review, we regard FoxO1 activation as detrimental, promote high plasma glucose level, and induce insulin resistance. Indeed, many contrasting… Show more

“…The re-emergence of FOXO1 and PKD2 has caught our attention. The present study indicated that FOXO1 is strongly associated with glucose and lipid metabolism, especially in Diabetes mellitus type 2 [37] and NAFLD [38]. Furthermore, FOXO1 has been identi ed as a key factor in the progression of BC, serving as a bridge between miRNA, hormone, and drug therapy [39][40][41].…”

“…FoxO6 mediates insulin's effect on target genes and integrates insulin signaling with glucose and lipid metabolism in the liver. FoxO1 regulates transcriptional processes that promote gluconeogenesis and fatty acid oxidation, as well as the IGF1 and leptin pathways [42]. Another pathway is the mTOR signaling pathway, which regulates insulin secretion and β-cell metabolism and is associated with the development of insulin resistance and β-cell dysfunction in type 2 diabetes [43].…”

“…In the nervous system, FOXO1 can affect neuronal autophagy ( Castillo et al, 2013 ). On the other hand, FoxO1 is the main target of insulin signaling pathways, and as a result controls glucose metabolism ( Kousteni, 2011 ; Nathanael et al, 2022 ). These mechanisms suggest FOXO1 may have implications in progression of AD.…”
