The role of flavor and fragrance chemicals in TRPA1 (transient receptor potential cation channel, member A1) activity associated with allergies

Abstract: TRPA1 has been proposed to be associated with diverse sensory allergic reactions, including thermal (cold) nociception, hearing and allergic inflammatory conditions. Some naturally occurring compounds are known to activate TRPA1 by forming a Michael addition product with a cysteine residue of TRPA1 through covalent protein modification and, in consequence, to cause allergic reactions. The anti-allergic property of TRPA1 agonists may be due to the activation and subsequent desensitization of TRPA1 expressed in … Show more

“…Thymoquinone is also a significant constituent of M. fistulosa L. (Table 1). Interestingly, essential oils extracted from Nigella sativa (black cumin) seeds have therapeutic properties, including antitumor activity in multiple types of cancer [45,89], and Mihara et al proposed a mechanism of interaction between TRPA1 and thymoquinone via Michael addition [90].…”

“…Myrcene is a component of Thyme oil, some components of which are TRPV1 and TRPA1 activators (thymol, linalool) [48,49]. However, the target of Myrcene as an ion channel activator [50][51][52] has not been explicitly explored. We used wholecell patch-clamping to investigate Myrcene activation of TRPV1.…”

“…Fragrance chemicals like menthol, 1,8-Cineole, borneol, fenchyl alcohol etc. are associated with TRPA1 activation and play a key role in allergic reactions [16]. Cigarette smoke, wood smoke, diesel exhaust, and other combustion-derived particles activate TRPA1, causing irritation and inflammation in the respiratory tract [17,18].…”
