The role of CircRNA/miRNA/mRNA axis in breast cancer drug resistance

Abstract: Multidrug resistance is one of the major obstacles in the treatment of cancers. This undesirable feature increases the mortality rate of cancers, including breast cancer. Circular RNA (CircRNA)/microRNA (miRNA)/messenger RNA (mRNA) is one of the important axes with major roles in the promotion and resistance of breast cancer. This heterogeneous pathway includes mRNA of oncogenes and tumor suppressors, which are controlled by miRNAs and CircRNAs. Unfortunately, this network could be easily deregulated, resultin… Show more

“…In addition to the pathogenic changes brought on by alterations in the ceRNA networks, the effects on response and resistance to anticancer drugs have also been observed. CircRNA/miRNA/mRNA interactions have been implicated in resistance to paclitaxel (PAX), such as CircGFRA1/miR-361-5p/TLR4, Circ-0006528/miR-1299/CDK8, and Circ-ABCB10/let7a-5p/DUSP7, and 5-fluorouracil, such as CDR1as/miR-7/CCNE1 and CircFBXL5/miR-216b/HMGA2, as well as doxorubicin and tamoxifen, such as CircBMPR2/miR-553/USP4, hsa_circ_0025202/mir-182-5p/FOXO3, and miR-197-3p/HIPK3, and in various cancers, including breast cancer and TNBC [ 92 ]. Moreover, the lncRNA FTH1P3 promotes paclitaxel resistance in TNBC cells through a miR-206/ABCB1 axis [ 2 , 93 ].…”

“…CircRNAs, which have a highly stable thermodynamic structure, act on and modulate pro-or anti-apoptotic proteins has been stated. They regulate the apoptosis of drug-resistant cancer cells via these pathways emphasized [36]. For example, circAMOTL1 was seen to regulate the expression of AKT-related pro-apoptotic BAX and BAK and anti-apoptotic BCL-2 proteins in BC.…”

“…molecules have recently been found to attach to miRNAs as sponges, suppressing miRNA function. 19,20 miRNAs are a family of endogenously generated ncRNAs, approximately 22 nucleotides in length. They play regulatory roles in gene expression by binding to partially complementary target sequences in specific mRNAs, leading to their degradation and/or translational repression.…”

“…Despite being discovered in RNA viroids in the 1970s, circRNAs were overlooked in the past because they were considered byproducts of splicing events. These RNA molecules have recently been found to attach to miRNAs as sponges, suppressing miRNA function 19,20 …”
