Selenium‐biofortified broccoli (Brassica oleracea L. var. italica Plenck) is an important source of micronutrients in a healthy human diet. Therefore, studies are needed on the commercial production of selenium‐enriched broccoli. We analyzed the effect of the foliar application of sodium selenate and sodium selenite in doses of 50 and 100 g ha−1 on the growth, photosynthetic parameters, selenium accumulation, and commercial quality of broccoli in two cultivars (‘Belstar’ and ‘Legend’) used for fresh crops market grown under field conditions. The application of both doses of selenate and the highest dose of selenite induced a significant increase in the head fresh weight (FW) only in cultivar Belstar. However, the dry matter content of the heads was not significantly affected by selenium or the cultivar of broccoli. In addition, the increase in the water use efficiency, mainly due to the reduction of the transpiration rate, could explain the significant increase in the head FW without modifying the head dry matter content in Belstar. On the other hand, selenium treatments before harvest maintained the green color and did not affect the firmness and diameter of the heads at harvest. The selenium content of the heads increased markedly in a dose‐dependent manner in both cultivars, with both selenium treatments. The results of this study indicate that the response to selenium treatments differs between broccoli cultivars and that the use of both selenium salts in foliar form would be a good strategy to obtain selenium‐enriched broccoli, without affecting the commercial quality of the heads.

Brassinosteroids (BRs) represent a unique class of steroidal plant hormones that display pronounced growth‐promoting activity at very low concentrations. Although many efforts have been made to characterize the molecular basis of BR action, little is known about the mechanisms behind the growth‐promoting effect of BRs at protein level. Proteomic analysis of response to the steroid plant hormone 24‐epibrassinolide (EBR) in wheat seedling shoots (Triticum aestivum L.) was performed using two‐dimensional electrophoresis (2‐DE) and immunoblotting with highly specific antibodies (PY20) to phosphotyrosine. EBR‐modulated proteins and phosphotyrosine polypeptides were identified using MALDI‐TOF mass spectrometry. The study revealed that EBR‐stimulated growth of wheat seedlings was accompanied by changes in the content of multiple proteins as well as in tyrosine phosphorylation of numerous polypeptides. Among them, 22 differentially accumulated proteins and 13 phosphotyrosine proteins were identified. Based on their performed functions, the identified proteins are involved in physiological processes (photosynthesis, growth, energy and amino acid metabolism) closely associated with intensification of plant metabolism. The EBR‐induced changes in protein abundance and tyrosine phosphorylation profile may contribute to growth stimulation of wheat seedlings under the action of EBR. The obtained data suggest an important role for EBR in the activation of protein metabolism underlying fundamental physiological processes, including growth promotion.

BACKGROUNDAmong the environmental stressors, which oxidative action is currently widely studied, are mycotoxins. The aim of presented studies was to investigate the response of leaves of seedlings to zearalenone (ZEN) applied directly to leaves and to grains (indirectly) of tolerant and sensitive wheat cultivars.RESULTS Biochemical analyses of antioxidative activity were performed for chloroplasts and showed the similar decrease of this activity independently of plant sensitivity and the way of ZEN application. On the other hand, higher amounts of superoxide radicals (microscopic observation) were formed in leaves of plants grown from grains incubated in solution of ZEN and in sensitive variety. Electron paramagnetic resonance (EPR) studies showed that upon ZEN treatment larger amount of Mn - aqua complexes was formed in leaves of tolerant wheat cultivar than in those of sensitive one whereas the degradation of Fe-protein complexes occurred independently of the sensitivity of plant varieties. CONCLUSIONThe changes in the quantity of organic stable radicals, which were formed by stabilizing reactive oxygen species on biochemical macromolecules, pointed to the higher ability to their generation in leaf tissues subjected to foliar ZEN treatment indicating the important role of these radical species in protective mechanism mainly against direct toxin action. The way of activation the defense mechanisms depended on the method of applications toxin.
