Extracellular alkaline phosphatase enzyme activity (APA) is important for algal phosphorus (P) acquisition in P-limited freshwater ecosystems and is often used as an indicator of P deficiency. APA allows access to organic P (monophosphate esters), but the regulation of APA in response to availability of both PO 43) and organic P is poorly characterized. This study aimed to examine the regulation of APA in freshwater Cladophora-epiphyte assemblages in response to PO 43) and a hydrolyzable organic P source, and for the first time to apply enzyme linked fluorescence (ELF) to localize APA within freshwater macroalgal-epiphyte assemblages. In response to elevated PO 43) concentrations, a component of net APA was suppressed, but there was also a constitutive APA, which was maintained even after prolonged exposure to nearly 1,000 lM PO 43) and saturation of internal P pools. When supplied with organic glycerol P as the sole P source, the algae maintained APA in excess of needs for supplying PO 43) for uptake, resulting in PO 43) release into the medium. Constitutive APA may be adaptive to growth under chronic P limitation in oligotrophic freshwater habitats. Excess APA and release of PO 43) could benefit different algal and bacterial partners within assemblages. APA in both Cladophora sp. and epiphytic algae was localized with ELF only when ethanol fixation was omitted. In algal subsamples exposed to different P treatments, there was no correlation between bulk APA (using 4-methylumbelliferyl phosphate [MUP] substrate) and % cell labeling with ELF, suggesting that ELF labeling of APA was at best semiquantitative in the algal assemblages.

Extracellular phosphatases are an important part of the phosphorus cycle in aquatic environments. Phosphatase activity (PA) in plankton was studied in seven subtropical shallow lakes of different exploitation management and trophic status in the urban area of Wuhan City. Bulk PA was rather high (range 1.1-11 lmol l -1 h -1 ), although concentrations of soluble reactive phosphorus (SRP) were also high (range 27 lg P l -1 to *1.5 mg P l -1 ) in all lakes. Cell-associated extracellular PA in phytoplankton was detected using the fluorescence-labelled enzyme activity technique. Phytoplankton species partly contributed to the bulk PA. We found explicit differences in the presence of cell-associated phosphatase within the main phytoplankton groups; species belonging to Chlorophyta and Dinophyta were regularly phosphatase-positive, while Cyanophyta and Bacillariophyceae were phosphatasenegative in all but one case. Furthermore, there is a certain potential of extracellular phosphatases produced by heterotrophic nanoflagellates in most of the lakes. This new finding compromises the 'traditional' interpretation of bulk phosphatase data as being due to overall phytoplankton or bacterial P regeneration.

High bulk extracellular phosphatase activity (PA) suggested severe phosphorus (P) deficiency in plankton of three acidified mountain lakes in the Bohemian Forest. Bioavailability of P substantially differed among the lakes due to differences in their P loading, as well as in concentrations of aluminum (Al) and its species, and was accompanied by species-specific responses of phytoplankton. We combined the fluorescently labeled enzyme activity (FLEA) assay with image cytometry to measure cellspecific PA in natural populations of three dinophyte species, occurring in all the lakes throughout May-September 2007. The mean cell-specific PA varied among the lakes within one order of magnitude: 188-1,831 fmol AE cell )1 AE h )1 for Gymnodinium uberrimum (G. F. Allman) Kof. et Swezy, 21-150 fmol AE cell )1 AE h )1 for Gymnodinium sp., and 22-365 fmol AE cell )1 AE h )1 for Peridinium umbonatum F. Stein. To better compare cell-specific PA among the species of different size, the values were normalized per unit of cell biovolume (amol AE lm )3 AE h )1 ) for further statistical analysis. A step-forward selection identified concentrations of total and ionic Al together with pH as significant factors (P < 0.05, Monte Carlo permutation test), explaining cumulatively 57% of the total variability in cell-specific PA. However, this cell-specific PA showed an unexpected reverse trend compared to an overall gradient in P deficiency of the lake plankton. The autecological insight into dinophyte cell-specific PA therefore suggested other factors, such as light availability, mixotrophy, and ⁄ or zooplankton grazing, causing further PA variations among the acidified lakes.
