The role of arabidopsis AtFes1A in cytosolic Hsp70 stability and abiotic stress tolerance

Abstract: SUMMARYAtFes1A is induced by high temperatures, and encodes a protein containing the armadillo repeat motif. Little is known about its biological function, however. In this study, we observed an increased heat-sensitive phenotype in atfes1a mutants, suggesting the involvement of AtFes1A in acquired thermotolerance. We found that AtFes1A is cytosolic and associates with cytosolic Hsp70. Loss of AtFes1A leads to a selective reduction of cytosolic Hsp70 and a global increase in heat shock transcription. Thus, AtF… Show more

“…This has for example been shown to be the case in early defense response, where the E3 ubiquitin ligase CYS, MET, PRO, and GLY PROTEIN 1 (CMPG1, AT1G66160), which itself is a target of the UPS, gets stabilized and accumulates (80). This also applies to the heat shock protein HSP70, as cytosolic HSP70 associates with FES1A (AT3G09350), which is induced by high temperatures and prevents HSP70 degradation (81). The level of FES1A was increased after SylA treatment (Table I), as were transcript levels for HSP70 and FES1A.…”

“…Therefore enhanced thermotolerance can also lead to enhanced salt tolerance. The Arabidopsis AtFes1A protein is involved in thermotolerance by stabilizing Hsp70 proteins, as shown in atfes1a knock out mutants, which had increased salt sensitivity and reduced thermotolerance [49]. This susceptibility was accounted for by increased ubiquitination of Hsp70 proteins, which are involved in thermotolerance.…”

“…The enhanced expression of HSP70 was reported to assist in translocation, proteolysis, translation, folding, aggregation, and refolding of denatured proteins (Gorantla et al, 2007; Zhang et al, 2010), while a methionine-rich chloroplast HSP has been shown to protect the thermolabile photosystem II and, consequently the whole-chain electron transport during heat stress (Peet et al, 2002). HSP101 for example, appears not to be required for growth in normal conditions, but plays a major role in tolerance to severe heat stresses and protein oxidative protection in Arabidopsis (Queitsch et al, 2000).…”
