The RNA Binding Protein Nuclear Factor 90 Functions as Both a Positive and Negative Regulator of Gene Expression in Mammalian Cells

Abstract: Nuclear factor 90 (NF90) was originally isolated in a complex that binds to the antigen recognition response element (ARRE-2) present in the interleukin-2 promoter. To characterize the transcriptional properties of NF90 in mammalian cells, we examined its ability to modulate promoter function in cellular transfection assays. NF90-Gal4 fusion proteins inhibited transcription from the adenovirus major late promoter in a fashion that was dependent on Gal4 targeting. Conversely, NF90 activated the cytomegalovirus … Show more

“…The resulting peptides were analyzed by mass spectrometry (16), which identified the 80 -90-kDa protein as the short form of ILF3 (also called NF90, NFAR-1, MPP4, and DRBP76). ILF3 is a nuclear protein that contains two dsRBD and an RGG domain and that influences gene expression (11,(17)(18)(19)(20)(21)(22). Importantly, the RNA/protein complex was dissociated upon addition of anti-ILF3 antibodies whereas mock antibodies had no effect (Fig.…”

“…NF90 and its homologs selectively regulate gene expression in mammalian cells, both positively and negatively (Saunders et al 2001b;Krasnoselskaya-Riz et al 2002;Reichman et al 2002). The dsRNA-binding region is essential for these regulatory actions but the role of RNA has not been directly established.…”

“…1. Additionally, a common complex partner of NF45 and a known dsRNA binding protein [26][27][28], NF90, was predicted to be part of the mRNP complexes and was tested for its ability to bind PGHS-1 mRNA sequence. (Fig.…”

“…1) after affinity purification, were used for validation experiments. An additional protein, NF90, was included in the investigation since it is a common binding partner of NF45 and a known dsRNA binding protein [26][27][28]. The affinity purification results were further validated by immunoprecipitation assays using antibodies specific to nucleolin, NF45, NF90 and serpin B1 followed by RT-PCR experiments to detect PGHS-1 mRNA.…”
