The RipA and RipB Peptidoglycan Endopeptidases Are Individually Nonessential to Mycobacterium smegmatis

Abstract: Mycobacteria possess a series of Rip peptidoglycan endopeptidases that have been characterized in various levels of detail. The RipA and RipB proteins have been extensively studied and are DL-endopeptidases, and RipA has been considered essential to Mycobacterium smegmatis and Mycobacterium tuberculosis. We show here that the ripA and ripB genes are individually dispensable in M. smegmatis and that at least one of the genes must be expressed for viability. We characterized strains carrying inframe deletion mut… Show more

“…RipA and RipB are not essential in M. tuberculosis. A recent study in M. smegmatis demonstrated that ripA is not essential, provided that ripB, coding for another D,Lendopeptidase and located downstream of ripA within the same operon, is expressed (28). To examine whether ripA and ripB are essential in M. tuberculosis, we first generated a merodiploid strain by expressing the ripA and ripB genes from an integrated streptomycin resistance-conferring plasmid in the attL5 locus (WT::ripAB) ( Fig.…”

“…Here, we identified RipA-a peptidoglycan hydrolase (Hett et al, 2008;Martinelli & Pavelka, 2016)-as a MarP substrate. MarP cleaves RipA and this processing is essential for RipA's activity.…”

“…9 In M. tuberculosis, Rv1477 and Rv1478 are individually non-essential and the knock out phenotype leads to lysozyme sensitivity (decreased cell wall stability) and increased susceptibility to various antibiotics. 10 Both proteins encoded in the operon harbor a NlpC/P60 domain 11…”
