The Remarkable Effect of Potassium Iodide in Eosin and Rose Bengal Photodynamic Action against Salmonella Typhimurium and Staphylococcus aureus

Abstract: Antimicrobial photodynamic therapy (aPDT) has been shown as a promising technique to inactivate foodborne bacteria, without inducing the development of bacterial resistance. Knowing that addition of inorganic salts, such as potassium iodide (KI), can modulate the photodynamic action of the photosensitizer (PS), we report in this study the antimicrobial effect of eosin (EOS) and rose bengal (RB) combined with KI against Salmonella enterica serovar Typhimurium and Staphylococcus aureus. Additionally, the possibl… Show more

“…Indeed, we show that the presence of human plasma with its high HSA content can seriously interfere with aPDT using 1D9-700DX. However, this antioxidant activity can be completely overcome through the application of KI, a nontoxic salt that was previously shown to improve the aPDT efficacy in several models (42)(43)(44)(45). Importantly, KI has been approved by the US Food and Drug Administration for thyroid blocking in radiation emergencies, which underpins its therapeutic application potential.…”

“…The spectral emission of the LEDs system was obtained using a spectrofluorometer (Varian Cary Eclipse, San Diego, CA, USA). The absolute irradiance of the LEDs was evaluated with a Spectroradiometer USB2000 + RAD (OceanOptics, Winter Park, FL, USA) 19 .…”

“…To evaluate the potential of aPDT with NMB and MB in the photo-inactivation of Psa and to select the best photo-inactivation conditions to be used in the pollen disinfection, in vitro assays were performed. The first experiments in vitro were performed in phosphate-buffered saline (PBS), using NMB at different concentrations (1.0, 2.5, and 5.0 μM), without and with KI addition (an aPDT potentiator), selected according to our previous studies [ 26 , 27 ] at a concentration of 100 mM. In a second step, the best in vitro aPDT conditions established for NMB were extended to MB (concentration 5.0 μM) and the assays were performed with and without the addition of 100 mM of KI.…”
