The relationship between expression of PD-L1 and HIF-1α in glioma cells under hypoxia

Abstract: Hypoxia inducible factor-1α (HIF-1α) up-regulates the expression of programmed death ligand-1 (PD-L1) in some extracranial malignancies. However, whether it could increase PD-L1 expression in intracranial tumor is still unknown. Here, we explored the relationship between HIF-1α and PD-L1 expression in glioma, and investigated their clinical significance. In glioma patients, HIF-1α and PD-L1 were overexpressed in high grade glioma tissues and were significantly associated with poor survival. In glioma cells, PD… Show more

“…HIF-1 and, more importantly, HIF-2 are required for the maintenance of cancer stem cells (CSCs) that are capable of self-renewal, and for generating both CSCs and differentiated cells [63]. HIF-1 promotes the CSC phenotype, enhancing the expression of KLF4, MYC, OCT4, SOX2, and NANOG [64][65][66][67]. Glioblastoma stem cells (GSC) are also favored by the stabilization of the Notch intracellular domain by HIF-1α.…”

“…In parallel, the VEGF signaling is closely connected with hypoxia and the expression of HIF1A, and the significant upregulation of the HIF1A gene during storage may explain the increase in VEGF-A secretion [54]. Interestingly, the expression of PDL1 involved in the immunomodulatory activity of MSCs was significantly upregulated after storage, and the PDL1 expression can also be associated with the increased expression of HIF1A [55]. Conversely, the level of IL-6 in the conditioned medium decreased significantly after storage, which was accompanied by downregulation of the IL6 gene (Figures 3 and 4).…”

“…By preventing T-cell infiltration, the growth and invasiveness of GBM are supported [35]. As researched by Ding et al, in glioma cell lines, hypoxia upregulates PD-L1 expression via HIF-1α [36]. This is due to Semaphorin3A present on their surface that binds to its receptor on the BBB, the neu-ropilin1.…”
