Aditoprim (ADP) is a newly developed antibacterial agent in veterinary medicine. The metabolism and disposition of ADP in swine, broilers, carp and rats were investigated by using a radio tracer method combined with a radioactivity detector and a liquid chromatography/ion trap/time-of-flight mass spectrometry. After a single oral administration, more than 94% of the dose was recovered within 14 d in the four species. The urine excretion was dominant in swine and rats, making up 78% of the dose. N-monodesmethyl-ADP, N-didesmethyl-ADP, and 10 new metabolites were characterized. These metabolites were biotransformed from the process of demethylation, α-hydroxylation, N-oxidation, and NH2-glucuronidation. After an oral dose for 7 d, ADP-derived radioactivity was widely distributed in tissues, and high concentrations were especially observed in bile, liver, kidney, lung, and spleen. The radioactivity in the liver was eliminated much more slowly than in other tissues, with a half-life of 4.26, 3.38, 6.69, and 5.21 d in swine, broilers, carp, and rats, respectively. ADP, N-monodesmethyl-ADP, and N-didesmethyl-ADP were the major metabolites in edible tissues. Notably, ADP was detected with the highest concentration and the longest duration in these tissues. These findings indicated that ADP is the marker residue and the liver is the residue target tissue.

Traditional metal toxicity tests on organisms have mainly focused on continuous exposure at a fixed concentration. However, organisms are more likely exposed to pollutants intermittently in estuarine environments that are significantly impacted by anthropogenic activity. The present study examined whether different copper (Cu) exposure regimes at an equivalent dose can induce different metabolomics effects on the oysters. An estuarine oyster Crassostrea hongkongensis was exposed to Cu continuously or intermittently at an equal dose (time × concentration) for 6 wk. Continuous exposure regimes included 2 doses of 3.3 μg/L for 24 h and 20 μg/L for 24 h, with corresponding equal doses of 2 intermittent exposure regimes of 20 μg/L for 4 h and 120 μg/L for 4 h, respectively. Time-course measurements suggested that Cu bioaccumulation was comparable at equal low doses between the continuous regime (3.3 μg/L for 24 h) and the intermittent regime (20 μg/L for 4 h), but there was considerable difference for the high dose under different regimes. Nuclear magnetic resonance (NMR)-based metabolomics suggested that continuous and intermittent Cu exposures led to similar metabolite variation pattern in gills at an equal high dose, including decreased amino acids (e.g., aspartate, glycine, isoleucine, leucine, lysine, phenylalanine, threonine, and valine), lower energy-related compounds (e.g., adenosine triphosphate/adenosine diphosphate, acetate, citrate, and glycogen), and altered osmolytes (e.g., homarine and taurine). These biomarkers indicated disturbance of osmotic regulation and energy metabolism induced by Cu exposure regardless of regime. In addition, the 4-h intermittent Cu exposure resulted in slightly fewer adverse effects compared with the corresponding equal-dose continuous exposure. Oysters appeared to recover during the intervals of Cu exposure. The results indicated that metabolomic effects induced by Cu were more dose dependent than the Cu exposure regime. Background: Comprehensive understanding of plasma metabotype of diabetes (DB), coronary heart disease (CHD) and especially diabetes with coronary heart disease (CHDDB) has kept lacking. The metabolic fingerprints of CHD, DB and CHDDB were investigated to reveal their pathogenic mechanisms and interactions and identify their specific biomarkers.Methods: The plasma metabolomic differences and links of 15 DB, 15 CHD and 30 CHDDB patients and 15 matched healthy controls were investigated by NMR-based strategy. The univariate and multivariate statistical analyses-based pattern recognition was combined with network analysis to determine disease specific biomarkers and understand the corresponding pathogenic pathways.Results: A total of 17 metabolites related to the development of disease were identified by comparative metabolomic analysis, and 6 of them were shared by the three diseases. The metabolites involved in amino acid synthesis (valine, alanine, leucine, isoleucine, and N-acetyl-glycoprotein) were positively associated with CHD and CHDDB (Odds Ratios (OR) >1); The trimethylamine oxide, glycerol, lactose, indoleacetate and scyllo-inositol are closely related to the development of DB to CHDDB (OR>1), and indoleactate (OR: 1.06, 95% confidence interval (CI): 1.01-1.12) and lactose (OR: 2.46, 95% CI: 1.67-3.25) are particularly prominent in CHDDB. The three different multi-biomarker signatures could serve to distinguish between CHDDB, DB, and CHD. All diseases demonstrated the disturbed glycolysis/gluconeogenesis and amino acid biosynthesis pathway. Furthermore, inositol phosphate metabolism, tryptophan metabolism and microbiota metabolism play a major role in the development of CHDDB from DB.Conclusions: The disease-specific multi-biomarkers provide promising capability in auxiliary diagnosis of DB-related disease and follow-up of disease progression and treatment. The comparative metabolomics strategy of multi-diseases offers a comprehensive perspective in disease-specific markers and pathogenic pathways.
