The regulation of glucose transporter (GLUT1) expression by the RNA binding protein HuR

Abstract: HuR is a ligand for nuclear mRNAs containing adenylate-uridylate-rich (ARE) elements in the 3'-untranslated region. Once bound to the mRNA, HuR is recognized by adapter proteins that then facilitate nuclear export of the complex. In the cytosol, HuR is thought to function to control stability and translation of its ligand message. We have previously demonstrated that HuR is constitutively expressed in the 3T3-L1 cells and shuttles from the nucleus to the cytosol, but remains predominantly nuclear in the preadi… Show more

“…In the present study we focused on VEGF to confirm in vivo the pathway previously described in pericytes, however, we cannot exclude the involvement of other mRNAs that are targeted by HuR [60,61] and involved in angiogenesis as well, such as TNFα [62]. Even though we demonstrated the involvement of isoform beta of PKC, we cannot rule out the implication of other isoforms such as alpha and delta.…”

“…RNAs of three known targets of ELAVL1 regulation, Slc7a1, Gclc and Slc2a1 (Yaman et al 2002, Song et al 2005, Gantt et al 2006, which contain 3 0 UTR-like ARE sequences, all increased in expression at the blastocyst stage. Our data are consistent with detection of Gclc mRNA and protein only at the mouse blastocyst stage (Stover et al 2000).…”

“…Three RBP target mRNAs were selected, as all are identified as targets of ELAVL1 in other tissues (Yaman et al 2002, Song et al 2005, Gantt et al 2006. g-Glutamylcysteine ligase catalytic unit (GCLC) is a key enzyme involved in synthesis of glutathione, which reacts to cellular stresses and deactivates destructive reactive oxygen species (Meister 1994).…”

“…Studies are needed to define the cis and trans elements responsible for this inhibitory translational control. Gantt et al (21) showed that an RNA-binding protein (RBP), HuR, binds to glucose transporter 1 and leptin mRNA in adipocytes. We also detected HuR binding to leptin mRNA (our unpublished observation) and are presently examining whether this and other RBPs regulate leptin mRNA translation or stability and whether nutritional or hormonal status can affect their binding to leptin mRNA.…”
