The recognition of three different epitopes for the H-type 2 human blood group determinant by lections ofUlex europaeus, Galactia tenuiflora andPsophocarpus tetragonolobus (Winged Bean)

Abstract: The chemical mapping of the regions of H-type 2 human blood group-related trisaccharide (Fuc alpha (1-2)Gal beta (1-4)GlcNAc beta Me) that are recognized by three different lectins, the so-called epitopes, are reviewed together with an account of how and why oligosaccharides form specific complexes with proteins as presently viewed in this laboratory. The occasion is used to report the synthesis of the various mono-O-methyl derivatives of the above trisaccharide that were used in these investigations. Also, Fu… Show more

“…The a(1,2)-linked fucose transferred by FUT1, can be detected by the lectin UEA-I. 24 As shown in Figure 1, HepG2 cells are not stained with UEA-I (Fig. 1a, left panel), indicating that these cells do not express a(1,2)-fucosylated glycoconjugates and suggesting that they are deficient in FUT1 activity.…”

“…Although not without precedent (8), the use of fluoride ion to open the stannylene ring formed on reactions of 1 with di-tzbutyltin oxide is noteworthy. Except for the preparation of 47, bromide-ion catalyzed glycosylations (16,17) were used to establish the interunit a-D linkages of the various congeners of methyl a-isomaltoside (29).…”

“…Except for 6 and 12, the 2,3,4-trisubstituted derivatives of methyl a-D-glucopyranosides (17-21), reported in Scheme 2, were prepared as previously reported (8). All of the bromides reported in Scheme 3 were prepared from literature compounds.…”

“…5) establish the concept of differential conformer selection by lectins [Siebert et al, 1996;Poppe et al, 1997;Gilleron et al, 1998;von der Lieth et al, 1998;Harris et al, 1999]. Not only different sections of functional groups of a carbohydrate ligand are relevant for binding, as documented for example for plant, animal and bacterial galactoside-or plant blood group H (type 2)-trisaccharide-binding lectins by chemical mapping [Rivera-Sagredo et al, 1991;Du et al, 1994;Haataja et al, 1994;Solís et al, 1994;Solís and Díaz-Mauriño, 1997]. Importantly, each of the conformations in solution can be selected by different receptors as bioactive.…”
